Objective:To observe the effect on mRNA expression of TNF-α and IL-1β inin ischemic penumbra of cerebral infarction model rats treated by acupuncturing DU meridian acupoints; and to explore the mechanisms of protecting ischemic brain tissue by acupuncturing DU acupoints.Methods:Using the method of plugging wire into SD rats’middle cerebral artery to make cerebral infarction model which as experimental object. Those SD rats were divided into5groups including the narmal group,the sham-operated group, the model group, the acupuncture group and the compare group which has10SD rats in each group. The acupuncture group was treated by acupuncturing DU meridian acupoints and the compare group was treated by acupuncturing Yangming meridian acupoints. Then the changes of neurobehavioral scores were observed before and after acupuncture treatment, and the changes of the mRNA expression of TNF-α and IL-1βin ischemic penumbra of cerebral infarction model rats which were detected by Real-time PCR method were observed too.Results:(1)The effect on neurobehavioral score:The neurobehavioral score of both the acupuncture group and the comparative group are significantly improved before and after acupuncture treatment compared with the model group(P﹤0.05),and the effect on neurobehavioral score of the acupuncture group are more better than the compare group(P<0.05).(2) The effect on mRNA expression of TNF-α and IL-β:The mRNA expression of TNF-α and IL-1β in sham-operated rats’brain tissue are based on level expression. The mRNA expression of both in the model group rats’brain tissue are significantly higher expressed than in the normal(P<0.01).In the acupuncture group and the compare group rats’ brain tissue, the mRNA expression of both are lower than in the model group(P<0.01&P<0.05),however higher than in the normal group and in the sham operation group.Conclusion:There has more significantly effect to improve cerebralinfarction model rats’ neurological damage by acupuncturing DU meridian acupoints, and the acupuncture group can significantly reduce the mRNA expression of TNF-α and IL-1βin ischemic penumbra of cerebral infarction model rats than the compare group which acupuncturing Yangming meridian acupoints. Those maybe the mechanisms of restraining the inflammatory response of cerebral infarction by the method of acupuncturing DU meridian acupoints to reduce the excess mRNA expression of TNF-a and IL-1β in ischemic penumbra.
