| Objective: MiRNAs are a length of18~25nucleotide non-coding RNA,50%to60%of protein coding genes of the human subject to miRNA regulation, relies on itscomplex regulatory networks, also served an important role in hematopoietic function andthe formation of acute leukemia. This study researched expressions and their clinicalsignificance of miR-128, miR-17and miR-18a in pediatric ALL, to explore thesignificance between these miRNAs and ALL types, prognosis and relapse, looking fornew indicators of prognosis and relapse.Methods:63ALL cases, indluding48newly diagnosed,10remission,5relapse and10control. Extract the bone marrow or peripheral blood samples of total RNA and thenreverse transcription into cDNA by specific primers. Three miRNAs: miR-128, miR-17,miR-18a were detected by real-time fluorescence quantitative PCR. Expression ofmiRNAs was analyzed with comparative method in Ct value. Analyze the three miRNAexpressions of newly diagnosed, remission and relapse ALL, and in accordance with theclinical data for further analysis to explore the role of the three miRNAs in childhood ALL.Results: Expressions of miR-128, miR-17and miR-18a in newly diagnosed andrelapsed children were significant high, and remission after chemotherapy cases decreased.Combined with clinical data to do further analysis can found that miR-128, miR-17expression in T-ALL is much higher than the B-ALL, there was significant difference,With the rise of the degree of risk, miR-128expression was increased, miR-128expressionin standard risk, medium risk, high-risk group of were significantly different. MiR-17andmiR-18a in MLL gene rearrangement-positive cases were high expression, suggesting thatthey may play an important role in the incidence of MLL gene rearrangements inchildhood ALL.Conclusion: High expression of miR-128, miR-17and miR-18a in newly diagnosedand relapsed cases, and with the rise of the degree of risk, miR-128expression wasincreased, indicates that its expression may be related to the incidence of childhood ALL, and can be used as monitoring molecular markers of the effect of chemotherapy. MiR-128and miR-17expression in T-ALL is much higher than the B-ALL. MiR-17and miR-18a inMLL gene rearrangement-positive cases were high expression, indicates that theirexpression can provide a reference on prognosis, miR-17and miR-18a may be involved inthe formation of MLL gene rearrangement ALL, and affect its prognosis, indctes thatwhether it can be used to be molecular markers to judge high-risk ALL. |
