Objective: To investigate expression of Fas protein and FasL protein andmethylation of Fas gene in Esophageal squamous cell carcinoma tissues,Paracanceroustissues as well as in metastatic lymph nodes and non-metast atic lymph nodes. Analyze thecorrelation of Fas protein,FasL protein and methylation of Fas gene. Discuss the effectionthem may cause in development of Esophageal squamous cell carcinoma.Methods:54cases of esophageal squamous cell carcinoma tissues and correspond-ing paracancerous tissues were collected as well as35cases of metastatic lymph nodesand19cases of non-metastatic lymph nodes were collected from surgical patients.Fas andFasL protein expression of them were detected with immunohistochemical S-P method.Methylation of Fas gene of them were detected with methylation-specific PCR,MS-PCR.Analyze the correlation and difference among Fas protein,FasL protein,methylation of Fasgene and clinical-pathological data.Results:1.In tumor specimen the positive rate of Fas protein55.6%(30/54) is lowerthan85.2%(46/54) in corresponding paracancerous tissues (P<0.05). The positive rate ofdifferentiation of esophageal squamous cell carcinoma of Fas protein expression:highdifferentiation of84.2%(16/19),moderately differentiated of44.4%(12/27),25.0%(2/8) lowdifferentiation. Well-differentiated squamous cell carcinoma of the Fas-positive rate wassignificantly higher than in moderately differentiated or low differentiation (P<0.05).Different pathologic types of esophageal squamous cell carcinoma mdeulla, Tanumbrella,ulcers and narrowing between the various types of Fas protein positive expressionrate of59.3%(16/27),53.8%(7/13),50.0%(3/6) and50.0%(4/8)(P>0.05). Depth of invasion (T stage) of esophageal squamous cell Fas positive expression rate:T2of59.3%(16/27),T3of52.0%(13/25),T4of50.0%(1/2)(P>0.05);Lymph node metastasis in esophagealsquamous cell carcinoma of Fas protein positive expression rate was38.1%(8/21), withoutlymph node metastasis was66.7%(22/33)(P<0.05); Various clinical stages of esophagealsquamous cell carcinoma of Fas positive expression rate: Phase IIa61.9%(13/21), phaseIIb50.0%(5/10),phase III52.2%(12/23)(P>0.05).With no relations between sexuality,age(P>0.05).2. In tumor specimen the positive rate of FasL protein81.5%(44/54) is higherthan48.1%(26/54) in corresponding paracancerous tissues(P<0.05). FasL proteinexpression positive rate in esophageal squamous cell carcinoma of various degrees ofdifferentiation: well-differentiated57.9%(11/19),moderately92.6%(25/27),poorly differen-tiated100%(8/8), well-differentiated was lower than that in moderately or poorlydifferentiated(P<0.05); Different pathologic types of esophageal squamous cell carcinomamedulla, Tan umbrellas,ulcers and narrowing between the various types of FasL proteinpositive expression rate of81.5%(22/27),84.6%(11/13),83.3%(5/6) and75.0%(6/8)(P>0.05); Depth of invasion (T stage) in esophageal squamous cell carcinoma of FasL positiveexpression rate: T266.7%(18/27), T396.0%(24/25),T4100%(2/2),T2is lower thanT3,T4period(P<0.05); Lymph node metastasis in patients with esophageal squamous cellFasL protein positive expression rate was100%(21/21), without lymph node metastasiswas69.7%(23/33)(P<0.05),clinical staging of esophageal squamous cell carcinoma ofFasL positive expression rate:phase IIa71.4%(15/21),IIb and90.0%(9/10),III of87.0%(20/23)(P>0.05).With no relations between sexuality,age (P>0.05).3. In tumor specimenthe positive rate of methylation of Fas gene61.1%(33/54)is higher than22.2%(12/54) incorresponding paracancerous tissues (P<0.05). Different degree of differentiation ofesophageal squamous cell carcinoma of Fas gene methylation rate:well-differentiated63.2%(12/19),in the differentiation of59.3%(16/27),poorly differentiated62.5%(5/8)(P>0.05); the pathologic types of esophageal squamous cell carcinoma medulla,Tanumbrella,ulcers and narrowing between the various types of Fas gene methylation rateswere59.3%(16/27),61.5%(8/13),66.7%(4/6) and62.5%(5/8)(P>0.05);Depth of invasion (T stage)in esophageal squamous cell carcinoma of Fas gene methylation rate:T2of59.3%(16/27),T360.0%(15/25),T4100%(2/2)(P>0.05);Lymph node metastasis in patientswith esophageal squamous cell carcinoma of Fas gene methylation rate:61.9%(13/21),without lymph node metastasis:60.6%(20/33)(P>0.05);The clinical staging ofesophageal squamous cell carcinoma of the Fas gene methylation rates were as follows:Phase IIa61.9%(13/21), phase IIb60.0%(6/10), III60.9%(14/23)(P>0.05).With norelations between sexuality,age(P>0.05).4. In lymph nodes Fas expression rate33.3%(7/21) is lower than that in non-metastatic lymph nodes66.7%(22/33)(P<0.05);Metastasisgroup,lymph node FasL expression71.4%(15/21) higher than that in non-metastatic42.4%(14/33)(P<0.05);the transfer of lymph nodes Fas gene methylation rate57.1%(12/21), higher than the non-metastatic lymph nodes24.2%(8/33)(P<0.05).5.Thecorrelation index(r) between Fas and FasL protein expression of the correlation is r=-0.33,P<0.05;Fas and Fas gene methylation of the correlation coefficient r=-0.56,P<0.05;FasL and the Fas gene methylation correlation r=-0.09, P>0.05.Conclusions:1. The expression of Fas protein in esophageal squamous cellcarcinoma is down-regulation,FasL up-regulation.the Fas gene is Hypermethylation.theFas and FasL expression was negatively correlated,Fas expression was negativelycorrelated with the Fas gene hypermethylation performance.2. In metastastic lymph nodesFas is down-regulation,FasL up-regulation,and Fas gene is hypermethylation.3.Fasexpression has relations with the degree of differentiation and lymph node metastasis.Inwell differentiated Fas expression is higher than in poorly differentiated(P<0.05);in lymphnode metastasis Fas protein is lower than without lymph node metastasis(P<0.05). With norelations between sexuality,age pathological type,depth of invasion and clinical stage.4.FasL expression in esophageal squamous cell carcinoma has relations with the degree ofdifferentiation,lymph node metastasis,depth of invasion.well-differen tiated squamous cellcarcinoma of FasL expression is lower than in poorly differentiated(P<0.05);lymph nodemetastasis in esophageal squamous cell carcinoma of FasL protein was higher than thatwithout lymph node metastasis (P<0.05);T2period is less than T3,T4period(P<0.05). With no relations between sexuality,age,pathological type and clinical stage.5.The Fasgene methylation has no relations with sexuality,age,pathologic type, degree ofdifferentiation,depth of invasion,lymph node metastasis and clinical stage.
