Objective To study the correlation of SATB1gene and HER-2gene in human breastcancer tissues, and detect whether SATB1gene regulate HER-2gene in human breastcancer cell line MCF-7.Methods1. The expressions of SATB1and HER-2protein were studied in breastcancer samples by SP immunohistochemistry (IHC) analysis and to study the relationshipof SATB1and HER-2with clinical pathological parameters. SPSS18.0software was usedfor statistical analysis. Pearson χ2test was used for qualitative data. Spearman rankcorrelation was used for correlation between groups.2. The vectors pcDNA3.1-control,pcDNA3.1-SATB1were transfected into MCF-7cells using liposome transfection reagent,respectively.3. The transfected cells were selected in growth medium containing G418.G418-resistant cells were then subcloned with limited dilution.21days later, all individualG418-resistant clones were screened by RT-PCR and Western Blot to detect the geneexpression of SATB1and HER-2.Results1.The expression of SATB1in breast cancer was higher than that in adjacentnormal tissue, and the difference was statistically significant(χ2=21.987, p<0.001). Theexpression of HER-2in breast cancer were higher than that in adjacent normal tissue(χ2=14.644, p<0.001). The expression of SATB1had positive correlation with HER-2(rs=0.366,p=0.003).2. In a total of63breast cancer patients, age, pathological types, tumor size,lymph nodes status, ER and PR status were not associated with SATB1expression (p>0.05)while the histological grade was related to SATB1expression by univariateanalysis(p=0.037).3. The SATB1mRNA of transfected MCF-7cell withpcDNA3.1-SATB1vector was upregulated115.31±15.59times than that withpcDNA3.1-control vector. SATB1protein was expressed in transfected MCF-7cell withpcDNA3.1-SATB1vector.4. The HER-2mRNA of transfected MCF-7cell withpcDNA3.1-SATB1vector was correspondingly upregulated5.03±0.45times than thatwith pcDNA3.1-control vector. HER-2protein was expressed in transfected MCF-7cell with pcDNA3.1-SATB1vector.Conclusion In human breast cancer tissue samples, SATB1expression was correlatedwith HER-2. And one MCF-7cell line which overexpressed SATB1gene was established,in which SATB1expression can upregulate the HER-2expression.
