The Modulatory Effects Of Probiotics On IL-4、IL-8、IL-10and Ifn-γin Gastric Mucosa Of Helicobacter Pylori Infected C57BL/6Mice

Background and Objiective:Probiotics have shown beneficial effects in the treatment of Helicobacter pylori infection; however, the mechanisms behind such effects are not fully understood. In this study, we have investigated the immunomodulatory effects of probiotics in a mouse model of H. pylori infection.:To evaluate the therapeutic efficacy of probiotics for H.pylori-related gastritis in C57BL/6mouse model and to explore its therapeutic mechanism.Materials and methods:A total of32C57BL/6mouse were infected with H.pylori to establish the gastritis model, and then were randomly divided into4groups. Each group consist of8mice.The first group was treated with standard triple first-line therapy, the second group triple therapy combined with Lactobacillus and Bifidobacterium and Enterococcus, the third sodium chloride solution for14days. The rest Eight normal mice were taken as normal control group.Four weeks after the last treatment, the mice were all sacrificed. The infection of H.pylori in gastric mucosa of the mice was detected using rapid urease test, Giemsa staining and bacterial culture. The histopathological changes in the gastric mucosa of mice were estimated using hematoxylin-eosin staining. The ratio of Thl/Th2were detected by flow cytometry.Then the protein levels of IL-4,IL-8,IL-10,IFN-y in gastric mucosa were examined.Results:The eradication rates of H.pylori infection in mice treated with triple therapy combined with probotics was significantly increased compared with mice receiving only triple therapy(87.8%vs62.5%,P<0.05). The chronic inflammatory response of the mice infected with H.pylori was more serious than the mice receiving only triple therapy (2.33±0.45vs1.81+0.28,P<0.05),but both of them were more serious than the mice treated with triple therapy combined with probotics(1.07±0.26,P<0.05).There was significant difference between the mice treated with triple therapy combined with probotics and the normal control group.The ratio of Thl/Th2of the mice receiving triple therapy combined with probotics was lower than that of the mice receiving only triple therapy and the mice infected with H.pylori(1.78±0.12vs6.34±0.12,8.27±0.16,all P<0.05),but there was no significant difference compared with the normal control group(1.78±0.12vs2.06±0.14,P>0.05).In mice receiving triple therapy with probiotics,gastric neutrophil infiltration,IL-8, IFN-y were significantly decreased compared with the mice treated with triple therapy and the mice infected with H.pylori(74.56±5.84vs104.18±3.28,188.61±4.48;59.43±2.07vs107.32±3.06,169.82±3.81,all P<0.05),but there was significant difference compared with the normal control group(1.35±0.46,1.46±0.56,P<0.05),and the IL-8,IFN-y level in mice treated with triple therapy were significantly decreased compared with the mice infected with H.pylori(P<0.05),but there was significant difference compared with the normal control group(P<0.05).In mice receiving triple therapy with probiotics, IL-4, IL-10were significantly increased as compared with the mice receiving no probiotics,mice infected with H.pylori and the normal control group(43.46±3.62vs4.63±0.47,3.37±0.33,1.23±0.27;74.21±2.27vs5.24±0.23,4.51±0.42,4.73±1.42,all P<0.05), but there was no significant difference between the mice receiving no probiotics,mice infected with H.pylori and the normal control group (P>0.05).Conclusion:Standard PPI-based triple therapy supplementede with probiotics could be effective in increasing eradication rates of anti-H.pylori therapy.and reduced level of IL-8, IFN-y and neutrophil infiltration.Furthermore, the probiotics has the intrinsic ability to promote a Th2immune response through an increase in IL-10and IL-4...
