Objective To observe the expression of wnt inhibitor factor-1(WIF-1) and Wnt2b at the normal colorectal mucosa and colorectal adenocarcinoma, further to investigate the relation between the invasion and metastasis of adenocolorectal carcinoma and the expression of WIF-1and Wnt2b.Methods:①The expression of WIF-1and Wnt2b is detected in15cases of adjacent normal colorectal mucosa,18cases of adenomas and69cases of adenocolorectal carcinoma by immunhistochemical staining(streptavidine-peroxidase method).②RT-PCR is used to observe the expression of WIF-1mRNA and Wnt2b mRNA in5cases of the normal colorectal mucosa,6cases of adenomas and8cases of colorectal adenocarcinoma.Results:①The expression rate of WIF-1in colorectal adenocarcinoma is significantly lower than that in adjacent normal colorectal mucosa and adenomas. The expression rate of Wnt2b in colorectal adenocarcinoma is obviously higher than that in adjacent normal colorectal mucosa and adenomas.②The expression rate of WIF-1in colorectal adenocarcinoma do not correlate with the patients’age, gender, tumor size, Dukes stages, lymph node status and metastasis(P<0.05);The expression rate of Wnt2b in colorectal adenocarcinoma do not correlate with the patients’age, gender(P<0.05), but correlate with tumor size, Dukes stages, lymph node status and metastasis(P>0.05).③The expression rate of WIF-1and Wnt2b is negative related in the colorectal adenocarcinoma(r=-0.383, P<0.05).④WIF-1mRNA is expressed in adjacent normal colorectal mucosa, the expression in adenomas is reduced, and in colorectal adenocarcinoma it is reduced remarkably. But the expression of Wnt2b mRNA expression is gradually strengthened.Conclusion:①The expression of WIF-1periodically in normal colorectal, adenomas to colorectal adenocarcinoma is gradually weakened. It shows that WIF-1, as a Wnt signaling pathways cells antagonist, restrained Wnt signal pathway activation, thus inhibiting the tumor genesis of colorectal adenocarcinoma, and may play a role in the pathogenesis of colorectal adenocarcinoma.②The expression of Wnt2b plays a role in the pathogenesis of colorectal adenocarcinoma. Wnt2b expresses from normal colorectal tissues, adenomas to colorectal adenocarcinoma gradually strengthen, and the expression of Wnt2b is correlated with tumor genesis, invasion and metastasis; that Wnt2b may adjust the cell proliferation and differentiation in the formation process and plays a very important role.③Combined detection of the expression of WIF-1and Wnt2b can serve as a parameter for early diagnosis of colorectal adenocarcinoma and help for prevention and treatment of colorectal adenocarcinoma.
