Objective:The objective of study was to investgate the human papillomavirus (HPV) infection situation and analysis the age distribution of outpatients in LanZhou, and investigate the correlation of infection with the cervical lesion. Methods:(1) We first collected the sample of782outpatients in hospital in Lanzhou from Mar2010to Jul2011. Then21HPV-DNA types of the cervical exfoliative cytology sample were checked by DNA flow-through Hybridization genotyping technique. The age distribution of the HPV-DNA positive patients was analyzed.(2) The samples from the cytological abnormalities or high-risk (HR) HPV-positive patients were examined by colposcopy and cervical biopsy. The relationship between pathological situation and HR-DNA in cervical lesion was analized.(3) The expression of protein P16in the cervical tissue was examined by immunohistochemistry assay, and the correlation between the cervical lesion situation and the expression of P16was investigated. Results:(1) Among the782cases, the total positive rate was28.64%.17HPV genotypes from the total21HPV genotypes were found by Hubrimax. The most common genotypes were:HPV-16:7.67%(60/782), HPV-584.47%(35/782), HPV-68:3.07%(24/782), HPV-52:2.56%(20/782) and HPV-33:2.17%(17/782). In the positive cases, the single genotype and multiple genotype HPV infectious cases were18.54%(145/782) and5.50%(43/782) respectively. When analyzing the different age groups, we found that the peak age of HPV infection was between21and30years old.(2) With the aggravation of the cervical lesion situation, the HPV infection rate shows a upward tendency. In the45Normal or inflammatory cases,only16cases were HPV postive. However, the patients with cervical cancer, there were12cases in total14cases were postive.(3) With the aggravation of the cervical lesion situation, the expression of P16rised. In the45normal tissues, there are only ten cases were P16positive. However,18cases in32CIN Ⅰ cases,23cases in26CIN Ⅱ cases and14cases in14carcinoma tissues were P16positive. That is to say the expression of p16positively correlated with the HPV infection.(There was significant difference among the comparising groups, p<0.05) Conclusion:Our study shows that:(1) the postive rate of HPV infections in lanzhou city was28.64%;(2) with the aggravation of the cervical lesion situation, both the HPV infection rate and the expression of protein P16rised;(3) the combination of analyzing the degree of cervical lesion and the expression of P16could improve the the levels of diagnosis and treatment.
