| Purpose:to evaluate the efficacy and safety of the regime of FOLFOX6as the first line chemotherapy for elderly patients with advanced gastric cancer.Materials and Methods:44elderly patients((?)60years old) with histologically confirmed, advanced gastric cancer were treated with the chemotherapy of the FOLFOX6which consisted of intravenous infusion of Oxaliplatin(OXA)100mg/m2and Calcium Folinate (CF)400mg/m2dl plus5-Fluorouracil (5-FU)400mg/m2bolus dl and46-hour continuous intravenous infusion of5-FU2400mg/m2, every14days intervals.The statistical analysis was performed by SPSS16.0.Results:Forty-two patients were assessable for response.One achieved complete response (CR),nine patients demonstrated partial response (PR),and twenty-nine patients had stable disease. The overall response rate(ORR) was23.8%and the disease control rate (CR+PR+SD) was88.1%.The median time to progression and overall survival time were5.3months(95%CI:3.3-7.3months)and10.3months (95%CI:7.0-13.6months)respectively. In the patients without metastasis(n=21)had a significantly improved OS(17.0m Vs8.2m, p=0.048) as compared with metastasis. Forty-four patients were assessable for toxicity. The most severe hematologic toxicity was neutropenia (grade â…¢/â…£10.82%); the non-hematologic toxicity was hepatotoxicity (grade â…¢/â…£0.37%);none severe peripheral neuropathy was observed in our study, and there was no treatment related death.Conclusions:FOLFOX6regimen is effective and well tolerated in Chinese elderly patients with advanced gastric cancer. Further clinical trials are warranted. |
