| Background:Diabetes known for its dangerous complications, and the late complication-s is irreversible. Diabetic nephropathy lesions are minor, reasonable treatmentcan still reverse the pathological changes.Although renal biopsy can confirmthe diagnosis, but renal biopsy is invasive examination. So far, The earlydiagnosis of diabetic nephropathy is still a lack of a specific marker for simpleand effective. In recent years, genetic susceptibility to diabetic nephropathybecome a hot research. Genetic diagnosis in the future may become animportant indicator for early diagnosis of diabetic nephropathy. Currently, thereis fewer diabetic patients histone acetylation. To this end, the following studies:Objective:To observe the expression of HDAC2ã€HDAC3ã€HDAC8and HDAC9mRNA in the peripheral blood cells in the high glucose condition..Methods:In the experimental study, object of study were divided into two groups:50patients with T2DM diabetes group,50cases of healthy control group; Takethe peripheral blood and extraction of RNA,after reverse transcription reactionusing real-time polymerase chain reaction (Real time-PCR) detect T2DMHDAC genes mRNA expression. Results:Real time-PCR results showed that HDAC2ã€HDAC3ã€HDAC8andHDAC9mRNA expression in patients with T2DM were3.26±1.28-fold (P<0.01)ã€1.32±0.54-fold (P<0.05)ã€0.78±0.26-fold (P>0.05)and1.57±0.61-fold(P<0.05) of the healthy control group. Conclusion:HDAC2expression was higher in diabetes than healthy people. The highlyexpressed HDAC2gene provides a theoretical basis to find new directions ofthe treatment and early diagnosis of the various complications of diabetic DMin clinical. |
