Objective To investigate the effects of thrombopoietin(TPO) and tansforming growth factorβ1(TGFβ1) on the proliferation and differentiation of Dami Cell a human megakaryobiastieleukemia cell line.Methods1. By using CCK8assay the best stimulating concentration of TPO and the bestsuppression concentration of TGFβ1were detectedafter the DAMI cells cultivation in vitrowith different concentration of TPO and TGFβ1in the cultivation.2. Proliferation viability ofDAMI cells (trypan blue exclusion assay), the cell colony formation byAcetyl cholinesterase(AchE) colour and the expression of CD41+by fluorescence activated cellsorter were observed.Before the treatment of TPO Dami cells were cultured for12hours inTGFβ1.Results The stimulation is obvious in the concentration of TPO(250ng/ml).It’s opticaldensity (OD) was0.902±0.039F=50.250P<0.01by CCK8assay. The cell viability wasmaximum54.223±2.037%F=218.929P<0.01by using Trypan blue assay. When TGFβ1was used at the concentration of25ng/ml, the optical density (OD) was0.902±0.039F=50.250P<0.01and the cell viability was18.673±0.508%F=257.218P<0.01by using Trypanblue assay. After the cells were intervenedby TGFβ112hours, tpo was added. In the group ofTPO and low dose of TGFβ1the optical density (OD) was1.924±0.042(t=10.725P<0.01) byCCK8assay. In the group of TPO and high dose of TGFβ1the optical density (OD) was0.712±0.092(t=6.051P<0.01) by CCK8assay.Conclusion TPO and TGFβ1could affect proliferation and differentiation of megakaryocyte.TPO could counteract the effect of low dose TGFβ1on proliferation and differentiation ofmegakaryocyte.
