ObjectiveIncreasing number of Clinical oral implants in patients of diabetes with osteoporosis, Theexperimental by simvastatin on serum BGP and IL-6levels and bonemineral density(BMD) inSTZ-induced diabeticrats After planting. Provide the experimental basis for clinical use indiabetic patients with osteoporosis after planting.Methods36SD healthy adult male rats divided at random into T0group (normal plantinggroups,n=12) and T group, after rats in T group was intraperitoneal jected withstreptozotocin(STZ) to set up the diabetic rats model, two group rats were implanted withTitanium in the proximal tibial metaphysic.Then the T group was randomly splitted to T1group(Growing group of diabetic) and T2group (Simvastatin treatment group), and each grouphas12rats. After surgery,each rat of T0and T1was given1.5ml physiological saline every day;the rats of group T2Were given5mg/kg simvastatin per day for12weeks. level of BGP andIL-6in the serum were fmeasured by ELISA and the bone mineral density(BMD) of tibia wasmcasured by dual-energy X-ray absorptiometry. Decalcified specimens of hard tissue sectionswithout implants,routine HE staining and electron microscopy to observe the histologicalchanges around the implant.Results1.Serological test resultsAt the12th week, the serum levels of BGP: compared with group T0, T1group significantlylower, and the difference has statistical meaning(p=0.004<0.01), T2group tended to increase, thedifference was not statistical meaning(p>0.05), Compared with group T1, T2group significantlyincreased, and the difference has statistical meaning(p=0.001<0.01);the serum levels of IL-6:Compared with group T0, T1group significantly increased, and the difference has statisticalmeaning(p=0.001<0.01); T2group has a downward tended, the difference was not statistical meaning(p>0.05); Compared with group T1, T2group significantly decreased, and the differencehas statistical meaning(p=0.001<0.01).2.Bone quality measurementsAt the12th week,tibia bone mineral density levels: compared with group T0, T1and T2group significantly decreased, and the difference has statistical meaning(p=0.001,p=0.003<0.01);compared with group T1,T2group significantly increased, and the difference hasstatistical meaning(p=0.005<0.01).3. the serum levels of BGP and IL-6and the correlation between tibia BMDAt the12th week,positive correlation between serum BGP and BMD, correlation coefficientis0.486(p=0.016<0.05); serum IL-6was a significant negative correlation betweenBMD.correlation coefficient is-0.446(p=0.006<0.01).4.Around the implant and histological changesDecalcified specimens of hard tissue sections without implants,routine HE staining andelectron microscopy to observe the histological changes around the implant.In the T1group,osteoblast cells was significantly reduced in the peri-implant,Trabecular bone was sparse,thinning and fracture ststus;T2group with simvastatin, Osteoblasts increased significantly in theperi-implant,Trabecular bone are woven into the mesh, Higher than in T1group.Conclusion:Simvastatin can increase the serum levels of BGP and reduce the serum levels of IL-6in thediabetic rats After planting, and can increased the bone mineral density around the implant.through the application of simvastatin increased the number of osteoblasts, trabecular rich in theperi-implant. In the promotion of bone formation may exist at the same time can promote boneformation and inhibition of osteoclast in two ways.
