Objective:To observe2diabetic (DM) rat, that after inject with insulin, the expression of bone morphogenetic protein2(BMP-2). To investigate the expression and meaning of the morphogenetic protein-2that influenced of insulin in type2diabetic rat peri-implant bone.Methods:1.30type2diabetic rats(weight200-220g) were divided into3groups:normal group, diabetic group and diabetic+insulin group.2.Type2diabetic model rats were fed on high-glucose-high-fat diet and injected by low dose of streptozotocin (STZ). Implant model rats were implanted the titanium implant (1.3mm X5mm).3. Diabetic+insulin group inject with insulin everyday. And the diabetic group and normal group inject with equally physiological saline.4.12weeks later, blood glucose levels and weight of the rats were collected, X rayed. Then the rats were killed tibia and serum were collected. Immunohistochemical technique was used to assess the expression of BMP-2in bone tissue around the implant.Results:1. General observation12weeks later,3groups rats were all health and survival. Diabetic group were obviously thin, more diet and more urine. Diabetic+insulin group were obviously closed to the normal group. After Implant Surgery, plated area of3groups rats were all Healed well, Inflammation and infection were not find, Implant were not loose.2. Bone densitometryThe bone mineral of the diabetic group were obviously less than the other two groups. The difference have Statistically significant(p<0.05). Done mineral of the diabetic+insulin group slightly density were less than the normal group. The difference also have statistically significant(p<0.05).3. X-ray observationsThe normal and diabetic+insulin group were not find reduce bone density between implant and bone.Diabetic group’s bone mineral were somewhat less. Trabecular bone were sparse.4. Observed by light microscopyHE cells staining Results:To Observe all rat’s bone that between implant and bone. Bone continuity of Diabetic group were less than the other groups, can find little osteoblasts. Bone continuity of the normal and diabetic+insulin group were better, Osteoblasts were more.5. Immunohistochemical resultsThe BMP-2expression of normal and diabetic+insulin group were intense and general, BMP-2expression of Diabetic group were less.Conclusion:1. Diabetes can decrease the BMP-2expression of the implant around. So we can affect the bone formation around the implant through reduce the BMP-2expression.2. Insulin can improve morphogenetic protein-2in type2diabetic rat peri-implant bone., maybe can improve the bone formation around the implant.
