| objective To explore the effect of MMP-9 in guinea pig model with Emphysema and effect and mechanism of Budesonide with Bambuterol on Emphysema guinea pig models and MMP-9.Methods Thirty healthy guinea pigs were randomly divided into 5 groups:norm- al control group, Emphysema model group, Budesonide treated group, Bambuterol tr- eated group, and Budesonide plus Bambuterol treated group. Guinea pigs were expos- ed to cigarette smoke for 2 months. Expression of MMP were detected by immunohis- tochemical method and effect of Budesonide and Bambuterol were observed.Results1.In comparison with Emphysema model group,Budesonide treated group presented a decrease in the degree of inflammation. Budesonide plus Bambuterol treated group lower than Budesonide treated group.2.The comparison of Lung histomorphology: In comparison with normal control group, every model group had a significant decrease in MLI and increase in MAN. Budesonide treated group lower than Emphysema model group in MLI and higher in MAN. Budesonide plus Bambuterol treated group lower than Emphysema model group and Budesonide treated group in MLI and higher in MAN.3.MMP-9 in normal lung tissue was slightly positive. Expressions of MMP9 were significantly higher in Emphysema model group than those in normal control group. Expression was significantly decreased in both Budesonide treated group and Bude sonide plus Bambuterol treated group than that in Emphysema model group, but still higher than that of normal control group. Budesonide plus Bambuterol treated group lower than Budesonide treated group.Conclusion Elevated MMP-9 may increase the excelluar degradation, and contribute to the development of emphysema. Budesonide/Bambuterol combination treatment may decrease expressions of MMP-9 in guinea pigs with Emphysema. |
