Objective:To observe the changes of descent pathway on the ventral spinal cord injury rats model observed.Methods:28clean grade Female Sprague-Dawley rats were randomly divided into4groups0They are sham operation group (n=6)and three spinal cord injury groups (15g.30g.50g)(n=6-8). This study adopted self-designed experimental devices to construct standardized ventral spinal cord injury animal models through15,30,50g trans-versal compression on the spinal cord. In the sham operation group, instead of injurying only the vertebral arch and horizontal axon were removed.12hours、96hours after surgery, all the rats were injected BDA in the cortex by directed craniotomy.2weeks later,animals were sacrificed.Take out and stain the following segment of injury.Behavior tests(Improved Tarlov scores. Inclined plate experiment BBB scores) were carried out after surgery、According to the data using homogeneity of variance test results, select the application of two-way ANOVA or rank sum test for statistical analysis.Results:After nerve tracer BDA and immunohistochemistry, the positive neurons were stained brown, spinal cord injury group than the control group decreased significantly; As increasing the force,the number of positive neurons was significantly reduced. The integral optical density IOD in T12-L1plane of spinal cord injury groups’rats were significantly reduced(p<0.05); The modified Tarlov grading score, inclined plane test and BBB scores of the spinal cord injury groups’rats were significantly lower than the control group (p<0.05); As increasing the qulity of the weight,the ventral pulling force of the rats increasing, the lower limbs modified Tarlov grading score, the critical angle experiments in rats gradually ramp test, BBB score is decreasing oConclusions:(1)BDA nerve tracer can be a good display of neurons in the neural descent pathways of the SCI rats urinate. thus provides a reliable morphological evidence in the process of spinal cord injury and repair regeneration of pathway.(2)The modified BDA injection and staining techniques, can avoid the leakage and improve the imaging quality effectively;(3)The Clinical outcomes is related to the suffered power.After the ventral spinal cord injury, samples with more abnormal brain center downstream pathway tend to have more servere dysfunction.
