ObjectiveTo research the correlation between concentration of miRNA-21 and let-7a in serum and content of corresponding tissue. In the meantime to study the different expression of miRNA-21 and let-7a in gastric cancer patients before and after operation. At the same time to survey different expression of miRNA-21 and let-7a in serum in the control group, gastric ulcer, gastric inflammatory polyps, intestinal metaplasia and gastric cancer patients.Objects and methodsObjects:Collected 29 patients with gastric cancer as sample which included preoperative and postoperative peripheral blood, tumor and normal tissue. Blood and tissue sample of 25 cases of heathy control,25 patients with distal gastric ulcer,18 patients with gastric antrum inflammatory polyps and 23 patients with intestinal metaplasia in endoscopy center were collected in the meantime.Methods:Fluorescence quantitative PCR was used to detect expression of miRNA-21 and let-7 a in serum and corresponding tissue samples.Statistical analysis:SPSS 17.0 software was used for statistical analysis.Results1. MiRNA-21 in tissue and serum had relevance with rs= 0.826 (P=0.000), while let-7a in serum samples had significant correlation with that in mucosal with rs= 0.889(P= 0.000).2. MiRNA-21 in serum reduced observably after operation two mouth later with t=0.511(P=0.030), while let-7a increased with t=0.770(P=0.000).3. Difference of miRNA-21 in serum were statistically significant in 5 groups withχ2= 10.814, while difference miRNA-21 in normal control group with gastric ulcer, gastric inflammatory polyps, intestinal metaplasia and gastric cancer apart with Z=0.949,0.750,1.930,3.009. Difference of let-7a in serum were statistically significant in 5 groups withχ2= 26.460, while difference of let-7a in normal control group with gastric ulcer, gastric inflammatory polyps, intestinal metaplasia and gastric cancer apart with Z= 1.188,1.362,2.395,4.847.Conclusions1. Concentration of miRNA-21 and let-7a in serum and content of corresponding tissue have correlation respectively, and miRNA-21 and let-7a in serum have both changed after operation. They will become a new non-invasive technique in evaluating results of operations combined with other genomic or proteomic.2. MiRNA-21 in serum rise in gastric cancer patients compared with the control group, let-7a in serum of intestinal metaphases and gastric cancer have decreased with the control group. They will become a new non-invasive technique in diagnosis of gastric cancer combined with other genomic or proteomic.3. Concentration of miRNA-21 in serum of heathy control, benign gastric lesion had no difference, while let-7a in serum of heathy control and benign gastric lesion had no difference. We can make a conclusion:miRNA-21 and let-7a are meaningless to distinguish heathy control and benign gastric lesion.
