Mycoplasma genitalium(Mg)was first isolated from two men with non-gonococcal urethritis (NGU) in 1981.Studies of related has progressed, its unique genome,growth metabolic trait and association between non-gonococcal urthritis,chronic prostatitis,mucopurulent cervicitis ,pelvic inflammatory disease had been concerned with scholars. The studies recently indicated, Mg was associated with urethritis ,cervicitis and pelvic inflammatory infection ,and validated by animal test, but the studies of Mg infection prostatitis had not been reported in this region. Chronic non-bacterial prostatitis was genitourinary system diseases which the rate of development was higher at present. Although bacterial invasion was important cause of prostatitis, Whereas, patients had indefinite pathogens evidence, only depended on experiential treatment, and effect of treatment was not perfect , so nosogenesis which need rapidly find out was significant . The aim of the present assay was to explore the action of Mg infection in chronic non-bacterial prostatitis and the situation of epidemiology, and lay the foundation of clinical diagnosis and epidemiology investigation for Mg. We detected Mg by Culture and PCR methods. Prostate specimens of 487 patients with chronic prostatitis were isolated by Culture, in contrast 75 healthy men. Clinic specimens culture needed about 20 days, When culture medium from red to yellow and clear, We transplanted another medium as 1:10 with 0.45um little filtration membrane, the same as three times, Culture medium of colored as 1:10 were transplanted another liquid –solid medium and continued culture, when medium turned tiny red , observed bacterial shape to identify under convert microscopy .at the same time ,prostate specimens of two groups were extracted DNA ,we designed 16S-rRNA gene sequence and Mg-Pa gene sequence of Mycoplasma genitalium.16Sr-RNA gene sequence included all Mg gene sequence , and amplified gene sequence of conservative region of 16Sr-RNA with Mycoplasma, whereas, Mg-Pa was adhesin protein gene in bacterial end , was special and has features .At first ,16Sr-RNA primers of Mg were amplified , then , all positive template were confirmed using the Mg-Pa gene heminested PCR .The expected 341bp product was produced on amplification of material from 16Sr-RNA gene of Mycoplasma genitalium, and the expected 281bp product was produced on amplification of material from Mg-Pa gene of Mycoplasma genitalium. Contemporaryly, we designed positive (type strain),negative (buffer ) and other Mycoplasma (Uu,Mh type strain)comparison. Two sets of primers were selected for their ability to detect <10 to 50M. All positive confirmed by culture and PCR was estimated. Result of test indicate: (1) positive rate of Mg in the prostatic secretion from the 487 patients was 7.39% (36/487). Among the 36 positive cases, 13 cases had other pathogens, of which Ureaplasma Urealyticum (Uu) was the most. In the controls ,the positive rate of Mg was 1.33% (1/75).The differences between the two groups were statistically significant (x2=3.88 P<0.05).
