objective To investigate the relative expression of microRNA-146a (miRNA-146a,miR-146a) in peripheral blood mononuclear cells (PBMCs) of patients with Throid-associated ophthalmopathy (TAO)and its clinical signifycance,and to make a supplement to the etiological and pathological theories of TAO and suggest a promising future therapeutic strategy.Methods Twenty TAO patients, Twenty GD patients without ophthalmopathy and Twenty healthy controls matched with age and gender were enrolled in this study. Total miRNA was isolated from peripheral blood monocytes(PBMCs)of TAO patients, GD patients without ophthalmopathy and healthy volunteer donors by TRzol reagent. First,cDNA was reverse transcripted by using the stem-loop primer base on the miRNA. Then. the RT product-cDNA was amplified using quantitative real-time PCR reaction (qRT-PCR) by miR-146a PCR primer. The products were analyzed by DNA electrophoresis.Results The relative expressions of miR-146a in TAO patients and GD patients without ophthalmopathy were lower significantly than that of healthy controls(p<0.05),the ratio of expression reduction are98.02%for TAO patients and67.60%for GD patients without ophthalmopathy respectively. The relative expression of miR-146a in TAO patients was lower than that of GD patients without ophthalmopathy(P<0.05), the ratio of expression reduction is30.40%.Conclusion1.The stem-loop qRT-PCR based on the SYBR Green1was sensitive and specific enough to detect miR-146a.2.miR-146a was involved in the pathogenesis of TAO,thus, it may be a potential therapy target for TAO.
