Objective To measure the expression levels of microRNA(miR)-146 a of bone marrow mononuclear cells in systemic lupus erythematosus(SLE)patients with thrombocytopenia and analyze the correlation between the expression levels of miR-146 a with clinical features in SLE patients with thrombocytopenia.Methods From November 2015 to January 2017,22 SLE patients with thrombocytopenia,16 SLE patients without thrombocytopenia and 3 normal controls were recruited from the Department of Rheumatology and Immunology,the First Affiliated Hospital of Guangxi Medical University.The expression levels of mi R-146 a of bone marrow mononuclear cells in all SLE patients and normal controls were measured by real-time PCR.The clinical datas of SLE patients were collected which including sex,systemic lupus erythematosus disease activity index(SLEDAI),hemorrhage,serositis,arthritis,nervous system damage,heart damage,lung damage,kidney damage,gastrointestinal damage,hemoglobin levels,blood platelet count(BPC),white blood cell count,anti-nuclear antibody(ANA),complement(C)3,C4,anti-ribonucleoprotein(n RNP/sm),anti-double strand(ds)-DNA antibody,anticardiolipin antibody(ACA),anti-smith(sm)antibody antibodyand direct antiglobulin test(DAT).Results1.Expression levels of miR-146 a in bone marrow mononuclear cells between SLE patients and normal controls was not statistically significant(P>0.05).Expression levels of miR-146 a in bone marrow mononuclear cells between SLE patients with thrombocytopenia and SLE patients without thrombocytopenia was not statistically significant(P>0.05).2.Compared with SLE patients without thrombocytopenia,the ege of SLE patients with thrombocytopenia was older(P=0.040),but the other demographic characteristics and clinical features between the two groups were not statistically significant(P>0.05).3.Compared with SLE patients with thrombocytopenia and without heart damage,expression levels of miR-146 a in bone marrow mononuclear cells in SLE patients with thrombocytopenia and heart damage was down-regulated(P=0.040).Expression levels of miR-146 a in bone marrow mononuclear cells in SLE patients with thrombocytopenia between the other clinical features were not statistically significant(P>0.05).ConclusionsExpression levels of miR-146 a in bone marrow mononuclear cells is not associated with the pathogenesis of SLE and the pathogenesis of thrombocytopenia in SLE,but it is associated with heart damage in SLE with thrombocytopenia. |
