ObjectiveThe NPC cell lines with exogenous expression of CYB5R2 were established to study the function of CYB5R2 in nasopharyngeal carcinoma, and thereby the prospects for the development of early markers in diagnosis and prognosis and novel therapeutic strategies.MethodFirst we cloned CYB5R2 gene from the normal testis tissue, transfected the expression plasmid and control into NPC cell lines respectively. Then MTT assay, colony formation assay, cell invasion assay and tumorigenesis in nude mice were done to investigate the biological behaviors of the transfected cells.ResultThe NPC cell lines with exogenous expression of CYB5R2 were established. The capabilities of proliferation, invasion and tumorigenicity in nude mice were significantly decreased in the CYB5R2-transfected cells HONE 1, CNE2 than empty vector- transfected.ConclusionThis is the first study to confirm that CYB5R2 can inhibit the biological behaviors of NPC cells and CYB5R2 is a putative tumor suppressor gene.
