Objective:To investigate the expression of RUNX3and DNMT1in human gastric cancer, and to find out the clinical significance and the effect of Helicobacter pylori (HP) infection.Methods:Reverse Transcriptase-Polymerase Chain Reaction(RT-PCR) and immunohistochemical method were used to detect mRNA and protein expression of RUNX3and DNMT1in70cases of primary gastric carcinoma and70paired adjacent marginal normal gastric carcinoma tissues from surgery.Results:The expression of RUNX3in gastric cancer tissues was significantly decreased(0.5782±0.3470vs0.8062±0.4105), but that of DNMT1was obviously improved(1.7258±1.6361vs1.2015±0.3583). The depression of RUNX3was correlated with differentiation degree and Lymph node metastasis (P<0.05).In gastric cancer the expression of RUNX3in the Hp-positive group was significantly lower than the Hp negative group (P<0.05).Conclusion:RUNX3gene and DNMT1gene may be involved in the development of gastric cancer. To some extent, Hp infection may involved in inactivation of RUNX3gene.
