Huatanfang Prescription Effect On The Transforming Growth Factor β1(TGF-β1) In The Serum And Expression Of Nuclear Factor κB (NF-κB) In The Lung Tissue Of Chronic Asthmatic Rats

Objective: To investigate Huatanfang prescription (Wenyanghuatanfangprescription and Qingyanghuatanfang prescription) on the transforming growth factorβ1(TGF-β1) in the serum and expression of nuclear factor κB (NF-κB) in the lungtissue of chronic asthmatic rats and explore the mechanism preventing and treatingasthma with Huatanfang prescription in order to supply experiment data forHuatanfang.Methods: One hundred and thirty-five male Wistar rats were divided randomlyinto nine groups: Normal control group, Asthma model control group, Dexamethasonegroup, Qingyanghuatanfang prescription low dose group, Qingyanghuatanfangprescription middle dose group, Qingyanghuatanfang prescription high dose group,Wenyanghuatanfang prescription low dose group, Wenyanghuatanfang prescriptionmiddle dose group, Wenyanghuatanfang prescription high dose group, each group hadfifteen rats. By the way of being sensitized and long term inspired with ovalbumin, wecopy chronic asthma rats model. At the same time, each group was given relevant medicines andobserved about normal symptoms. At the end of the experiment, all the rats were killed. Weobserved the changes of lung histomorphology of rats of each group, and detected TGF-β1in theserum by enzyme-linked immunosorbent assay and expression of NF-κB in the lung byimmunohistochemistry. The data of NF-κB is quantitative image analysis.Experimental data were analyzed by spss15.0statistical software.Results: The chronic asthmatic rats model were successful. The normalsymptoms and the changes of lung histomorphology of Every treatment group is betterthan Asthma model control group. Compared to Normal control group, Asthma modelcontrol group was a large number of inflammatory cell infiltration, airway epithelialinjury and airway wall thickening. Pathological changes in the treatment groupcompared with the asthma model group, Qingyanghuatanfang prescription low group,Wenyanghuatanfang prescription middle group and Wenyanghuatanfang prescription high group are most clearly reduce. The serum TGF-β1of Asthma model group andtreatment groups were significantly higher than Normal control group (P<0.05); everytreatment group was significantly lower than Asthma control group (P<0.05);traditional chinese medicine group reduce the level of serum TGF-β1which is betterthan Dexamethasone group (P<0.05), Qingyanghuatanfang prescription middle groupand Wenyanghuaranfang prescription high group are the most obvious. About theexpression of NF-κB in lung tissue, Asthma model control group was significantlyhigher than Normal control group; each treatment group was significantly lower thanAsthma model control group (P<0.05), but significantly higher than Normal controlgroup (P<0.01); compared with Dexamethasone group, Wenyanghuatanfangprescription middle and high group and Qingyanghuatanfang prescription low groupwere no significantly different (P>0.05), other traditional chinese medicine groupwere significantly higher than Dexamethasone group (P<0.05); the expression ofNF-κB of Wenyanghuatanfang prescription high group is lowest which is significantlydifferent with Qingyanghuatanfang prescription high group (P<0.05).Conclusion:1The experimental results suggest that Huatanfang prescription caneffectively alleviate the clinical symptoms and lung histomorphology changes ofchronic asthmatic rats, and its mechanism is to: reduce the NF-κB expression andserum TGF-β1content.2Different therapies about Huatanfang prescription both canreduce the expression of NF-κB and serum TGF-β1level, but there are differences interms of dose, which may be because the experimental model does not have the“syndrome” feature on.
