Objectv To investigate the expression of microRNA-21(miR-21)、microRNA-375(miR-375) in gastric cancer (GC) and corresponding normal-gastric epithelial tissues,detectits relationship with clinicopathological features. Analysis expression of Spry2、AEG-1inthe gastric cancer and corresponding normal-gastric epithelial tissues.The relationshipbetween the expression of miR-21or Spry2and clinical pathologic parameters in thegastric cancer was analysed.The relationship between the expression of miR-375or AEG-1and clinical pathologic parameters in the gastric cancer was also analysed.Methods TheqRT-PCR method was used to assess the abundance of miR-21、miR-375in50primary GCspecimens which were diagnosed and operated in frist affiliate Hospital of suzhouuniversity between2008and2009, in comparison with the corresponding adjacentnontumor tissues.The relationship between the expression of miRNAs andclinicopathological features was analyzed. Immunohistochemical staining to detectexpression of Spry2、AEG-1protein in the gastric cancer and corresponding gastricepithelial tissues,The relationship between the expression of miR-21or Spry2and clinicalpathologic parameters in the gastric cancer was analysed.The relationship between theexpression of miR-375or AEG-1and clinical pathologic parameters in the gastric cancerwas also analysed.Results (1)The expression of miR-21was significantly elevated ingastric cancers compared with their nontumor counterparts from patients undergoinggastric resection.The expression of miR-375was significantly decreased in gastric cancerscompared with their nontumor counterparts from patients undergoing gastric resection.(2)No significant relationship was found between the expression of miR-21/miR-375andclinicopathological features.(3) There was negative relationship between miR-21and Spry2mRNA in gastric cancer(r=-0.387, p=0.034).There was negative relationshipbetween miR-375and AEG-1mRNA in gastric cancer(r=-0.369,p=0.045).(4)Expressionof Spry2protein is significantly higher in nontumor counterparts epithelial tissues than ingastric cancer tissues.Expression of AEG-1protein is significantly lower in nontumorcounterparts epithelial tissues than in gastric cancer tissues(.5)Expression of Spry2mRNArelated with lymph node metastasis in gastric cancer.Expression of Spry2protein relatedwith lymph node metastasis in gastric cancer.Expression of AEG-1protein related withlymph node metastasis、tumor size in gastric cancer.There was no relationship betweenExpression of AEG-1mRNA and clinicopathological factor in gastric cancer.(6)There wasnegative relationship between miR-21and Spry2proteins in gastric cancer(r=-0.398,p=0.030,P<0.05).There was no relationship between miR-375and AEG-1proteins ingastric cancer(r=-0.313, p=0.128).Conclusion miR-21expression was significantlyelevated in gastric cancers,it is possible that miR-21play significant role in gastriccarcinogenesis through the control of Spry2.miR-375expression was significantlydecreased in gastric cancers,It is possible that miR-375play significant role in gastriccarcinogenesis through the control of AEG-1.
