The Cytology Research Of¹⁸F-FDG And¹⁸F-FLT To Evaluate The Early Curative Effect Of Chemotherapy

Purpose:To use18F-FDG and18F-FLT to evaluation early response of chemotherapy on colon cancer cell HCT-116.Methods:Measure cell proliferation inhibition rate by CCK-8method after24h treated by different concentrations of Oxaliplatin,and work out the IC20、IC50、IC70;18F-FDG and18F-FLT uptake and uptake inhibition rates were measured after24h or48h treated by Oxaliplatin in different dose,measure the quantity of lactate dehydrogenase and glucose before and after treated,and measure the degree of cellular apoptosis and cell cycle by flow cytometry.Results:Cell proliferation inhibition rates were (75.15±9.06)%、(64.24±7.15)%、(41.21±5.51)%、(32.73±4.64)%、(24.85±7.76)%、(18.18±3.34)%(13.94±1.85)%(9.10±1.85)%by CCK-8at different dose of Oxaliplatin after24h and IC20、IC50、 IC70were3.33μg/ml、103.33μg/ml、170.00μg/ml.After24h and48h treated by Oxaliplatin in different dose,the uptake rates of18F-FDG were (40.79±0.82)%、(32.08±1.22)%、(16.52±1.43)%and (24.33±1.34)%、(12.37±1.12)%、(5.63±0.64)%,the uptake inhibition rates were (17.93±0.92)%、(35.45±1.67)%、(66.76±0.67)%and(50.70±1.49)%(74.93±1.16)%(88.59±1.19)%; the uptake rates of18F-FLT were (25.23±0.81)%、(20.03±0.94)%、(11.72±0.91)%and(19.45±0.82)%、(11.39±0.96)%、(4.62±0.84)%,the uptake inhibition rates were (17.60±4.82)%、(34.58±0.76)%、(61.72±0.25)%and (40.65±2.13)%、(65.24±5.16)%、(85.90±3.02)%. With the increasing of the drug dose,18F-FDG and18F-FLT uptake declines, uptake inhibition rates rises, and the difference was statistically significant(P<0.05).After24h,the glucose and lactate dehydrogenase content in culture solution were (4.43±2.33) mmol/L (6.59±0.51) mmol/L (8.35±0.41) mmol and (55.41±5.49) U/L,(79.43±8.65) U/L,(103.34±7.71) U/L;early apoptosis rate of HCT116cell were (8.2±0.21)%,(11.2±1.02)%,(15.8±0.56)%. The early cell apoptosis rate and the content of LDH in culture solution rises with the increasing of the drug dose,and the content of glucose declines.And the difference was statistically significant(P<0.05).Conclusion:18F-FDG and18F-FLT is expected to reflect the inhibitory effect of chemotherapy on colon cancer as early as24h.
