Objective:To examine the chang of peripheral blood neutrophils (PMNs) proteomics in rats with severe acute pancreatitis(SAP) induced by sodium taurocholate and the role of interleukin-18(IL-18)、Nuclear factor-kappa B (NF-κB) activation in apoptosis of peripheral blood neutrophils in rats with severe acute pancreatitis (SAP).Methods:Sixty SD rats were randomly divided into three groups:ANP group and sham operation (SO) group. At3,6and12h after ANP induction, blood samples were collected from the inferior vena cava to measure serum amylase and IL-18; PMNs were separated to detect their apoptosis and the levels of NF-κB P65in nuclear of PMN; differential protein analysis in PMNs was performed by Label free quantification proteomics in ANP group and SO group; and pancreatic specimens were taken for pathological examination.Reasults:①PMN apoptosis rate decreased remarkably in the ANP group than in the SO group, and the decrease became more obvious over time (P<0.01).②Serum levels of IL-18in the ANP group increased significantly compared with those in the SO group (P<0.01).③The level of nuclear factor-KB P65in nuclear of Peripheral neutrophil in ANP group increased significantly compared with that in SO group; There was a negative correlation between the apoptosis rate of PMNs and serum levels of IL-18and the expression of NF-κB P65in SAP rats (r=-0.78, P<0.01),(r=-0.742, P<0.01);④In 12h time point,320differentially expressed proteins (219upregulation proteins and101downregulation proteins) were screened by Label free quantification proteomics between ANP group and SO group (P<0.05), there are four proteins associate with PMNs apoptosis in significant differential protein.Conclusions:Compared to control rats, PMN apoptosis was obviously delayed, and serum levels of IL-18remarkably increased in ANP rats and the expression of NF-κB is remarkably increased, indicating that IL-18and NF-κB may plays an important role in peripheral neutrophil apoptosis in SAP; the proteomics of peripheral blood neutrophils of rats alters in the SAP and found four PMN apoptosis-related differential protein.
