Objective Through the comparison of acute hemorrhagic necrotic pancreatitis in mice before and after the depletion of neutrophils and lung damage levels to judge the influence of the neutrophils in necrotizing pancreatitis and the lung injury.Methods 60 healthy male ICR mice by weight were randomly divided into four groups:normal control group (N, n= 15), pancreatitis group (P, n= 15), the removal of neutrophils group (T, n= 15) and the depletion of neutrophils pancreatitis group (TP, n= 15).In N group, it is only injected with saline solution.In P group, injected the taurocholic acid sodium to the pancreatic duct contraorbitally for building in the acute hemorrhagic necrotic pancreatitis mice model.In Group T we injectd the monoclonal antibodies Ly6G-1a8 to the mices’tail vein to make deleption of neutrophils in mice model.In TP group we build in pancreatitis model about 24 h after injection of antibodies. In each group after 24 h and before the injection of antibodies, we makes blood neutrophils count and ratio collected from the eye cein respectively.Each group after 24 h of building pancreatitis in mice is to be put to death.Observing mortality of each group mice, the injury of lung and pancreas, lung and pancreas tissue pathology HE staining and pathological grade.Result 1) The removal of neutrophils in each group by monoclonal antibody Ly6G-1 a8 were above 90%.T and TP group before and after dosing neutrophils differences have statistically significant (P< 0.01).2) In N group no mice died, mortality is O.In T group one mice died, the death rate is 6.7%.In P group five mice died, mortality is 33.3%.In TP group two mice died, mortality is 13.3%.Obviously pancreatitis group death mortality are highest in mice, and the death mortality which removed the neutrophils in pancreatitis mice is lower than mice did not remove the neutrophils.3) The pancreas and lung tissue pathology change in N and T group is not obvious.P group and TP group is typically seen the pathological changes in the lens, but TP group is lighter than P group. T and TP group shows different degree of lung pathological changes of typical ALI, but to a lesser in TP group.4) In acute pancreatitis group mice lung wet dry weight ratio, lung and pancreatic pathology score significantly higher than the removal of neutrophils pancreatitis group (p<0.01) (3.70±0.46,4.8±1.2,8.8±1.lvs 2.75±0.36,2.1±0.6,7.2±0.9).Oxygenation index is lower than the removal of neutrophils pancreatitis group p<0.05 (462.57± 21.56 vs 491.53±9.28). The lung wet weight ratio、lung and pancreas pathology score in removal of neutrophils group and normal control group of mice is no significant difference observed in the oxygenation index statistically significant (p> 0.05).Conclusion The effect the monoclonal antibody Ly6G-1a8 for neutrophil depletion is obvious, The clearance rate were above 90%.After the neutrophil depletion in mice pancreatitis and lung injury in mice could be alleviated, but it can’t prevent pancreatitis and the lung injury, therefore neutrophils is the acceerative role in the process of the development of pancreatitis complicated with lung injury.
