Age-related cataract is the most common type of cataract. At present, thereare many researches scholars think that age-related cataract is protein structurechange disease, lens protein denaturation and irreversible the protease lens onabnormal decomposition. In this paper the proteins and enzymes from the lenschange the Angle discusses its and age-related cataract incidence relation.Alpha-crystallin as the highest component in lens, its molecular chaperoneactivity plays an extremely important role in a person’s life, two ofthe α-crystallin proteinsubunit has the molecular chaperone activity. αB-crystallin is widely expressed in a variety of tissues and cells, and the highestexpression level is in the lens and striated muscle (myocardium and skeletalmuscle). In the lens, αB-crystallin and αA-crystallin are in the way of1:3pattern consisting of tetramer, exerts their biological effects. The basic functionof αB-crystallin is to recognizes and combined with the aggregation propensityof non-natural proteins or protein folding process of intermediate products,stable target protein conformation, prevent the formation of irreversibleaggregation of the protein product. Therefore, αB-crystallin, to the stability ofquantity and quality of intracellular protein and the stability of the intracellularenvironment, plays a very important role. In recent years, we found thatαB-crystallin involved in a variety of cell physiology and the stress process,and showed some kind of anti-organization function to injury of cells andtissues.Objective: The present study is to investigate the expression of aB-crys-tallin in normal lens of rat of different ages,and to explore the relationshipsbetween the expression of aB-crystallin and age-related cataract.Methods: Lenses were obtained from general Wistar rats of3,6,9,12 weeks old(10lenses) and homogenized with different concentrations ofstandard bovine serum. The proteins were resolved using SDS-PAGE gelelectrophoresis, Total concentration of aB-crystallin in lenses from rats ofdifferent ages were detected by the way of Western blot.Results: aB-crystallin was detected at a molecular weight of23.5kDa.The total concentration of aB-crystallin in general Wistar rats lenses decreasedobviously with the growth of age: to3-weeks of age, the level of proteinconcentration of6,9,12-weeks of age were reduced respectively to the levelof90.87%,83.40%,71.64%.Conclusion: aB-crystallin may be involved in the alteration of crystallinduring the development of lenses, and it may play an important role in theformation and aggravation of age-related cataract.
