Objective Studying lymphocytes DNA damage of workers who are exposed to Hexyl hydride using flow cytometry assay recognizing yH2AX, to explore the genetic toxicity of Hexyl hydride. Examining the neurobehavioral function of workers who exposed to Hexyl hydride using neurobehavioral test softwar e which Chinese computerized,to Explore and validate the possibility of nervous system damage caused by Hexyl hydride of low concentrations. Methods The peripheral blood lymphocytes were obtained from33workers who are occupationally exposed to Hexyl hydride(exposed group) and17workers who are not exposed to Hexyl hydride in the same factory(inner control) and52some residents who have not been occupationally exposed to adverse factors(external control).Flow cytometer assay detects DNA damage of the lymphocytes. At the same time,32workers exposed to Hexyl hydride(exposed group) and32workers not exposed to Hexyl hydridecontrol group) were selected from the same factory, who taking part neurobehavioral test. Results The mean difference of DNA damage rate of exposed group, inner control group and external control group was statistically significant (P<0.001). DNA damage rate (%) of the exposed group (14.30±15.08)was significantly higher than the inner control group of workers (6.01±3.25) and external control groups of island residents (0.32±0.45).The DNA damage of inner control is statistically higher than the external control (P<0.01or P<0.05). The geometric mean of fluorescence intensity of exposed group, inner control group and external control group was statistically different (P<0.01). The geometric mean fluorescence intensity of the workers in the exposed group (1.51±0.87) was significantly higher than the inner control group of workers (1.04±0.23) and external control groups (0.73±0.28).The geometric mean fluorescence intensity of inner control is statistically higher than the external control (P<0.01or P<0.05). Neurobehavioral test result showing, the scores of negative emotions of exposed group(tension-anxiety,depression-depression,anger-hostility,fatigue-inertia,stumbl e-confusion) is statistically higher than control group (P<0.05). Operating capacity such as average accuracy of mental arithmetic, mental arithmetic average index, average accuracy of listening digit span, listening digit span average index, average index of curve consistent, average accuracy of curve consistent and average accuracy of target tracking,of exposure group is statistically lower than control group (P<0.05). Conclusion Hexyl hydride can cause DNA damage, meaning genetic toxicity of Hexyl hydride.γH2AX flow cytometer assay can be a sensitive, objective and effective method to detecting DNA damage of peripheral blood lymphocytes. Chronic exposure in Hexyl hydride can cause damage on neurobehavioral function.
