In this paper, the influence of sinomenin and resveratrol on the human lymphocyte DNA, the protection effect of resveratrol and baicalin against human peripheral lymphocyte DNA damage induced by hydrogen peroxide(H2O2) and ethanol in vitro, were studied by the method of Comet Assay.The results showed:1. H2O2(10μM, 20μM, 30μM, 40μM and 50μM, 1h) promoted DNA damage of human peripheral lymphocyte in vitro in a dose-dependent manner.2. Ethanl(20mM, 30mM, 40mM and 50mM, 1h) increased DNA damage of human peripheral lymphocyte in vitro in a dose-dependent manner.3. Sinomenin(0.4μg/ml, 4μg/ml, 40μg/ml and 400μg/ml, 1h) enhenced DNA damage of human peripheral lymphocyte in vitro in a dose-dependent manner.4. Resveratrol(10μM, 20μM, 30μM and 40μM, 5h) did not show DNA damage effect on the human peripheral lymphocyte in vitro, but resveratrol(30μM, 5h; 40μM, 1h, 3h, 5h) could attenuate H2O2 induced lymphocyte DNA damage. The resveratrol(10μM, 5h; 20μM, 30μM, 3h, 5h;, 40μM, 1h, 3h, 5h) could attenuate ethanol induced DNA damage.5. Baicalin(20μg/ml, 3h, 5h; 25μg/ml, 1h, 3h, 5h) could attenuate H2O2 induced lymphocyte DNA damage; baicalin(20μg/ml, 25μg/ml, 5h) could attenuate ethanol induced lymphocyte DNA damage.With the precise and reliable data, this research illuminated the DNA damage and protection effect of natural products on the human peripheral lymphocyte. In addition, a new method was set up to detect the DNA damage and protection effect of herb compounds on the human peripheral lymphocyte. Sinomenin exhibited possible DNA damage potentiality on the human peripheral lymphocyte; baicalin and resveratrol could attenuate human peripheral lymphocyte DNA damage induced by H2O2 and ethanol in vitro. These findings should be emphasized in the genotoxic research of natural products. A further research is needed to explain the mechanism of DNA damage and protection effect.
