Background&Objective: In recent years, many scholars have started in-depth studyof keloid and have paid attention more on study of different parts of keloid, and expectednew findings in keloid neoplastic growth. Osteopontin is a calcium-binding phosphorylatedglycoprotein with a variety of biological activity. It is secreted by mesenchymal cells inextracellular matrix. It has tissue specificity and cell specificity and is regulated by a varietyof cytokines through the appropriate pathways. The existing experiments has confirmed thatOPN is involved in immune regulation, ischemia–reperfusion injury, transmission ofinformation between cells and gene expression, cell chemotaxis, cell adhesion, inhibition ofapoptosis, promotion in tumor cells growth development and metastasis, wound healing,angiogenesis, progression of fibrotic disease. A variety of biological activities have beenconfirmed in the occurrence and development of keloid. The studies have showed that OPNmay accelerate vascular repair through activating transcription factor-4to enhance vascularendothelial cell growth factors expression and stimulates endothelial cell adhesion andmigration. It also has relationship between tumor invasion and metastasis by participating intumor angiogenesis. Therefore, we assumed to detect the expression of OPN, VEGF andATF-4in central and margins of keloid and compare with normal skin usingimmunohistochemical staining techniques, then to explore OPN and VEGF, and ATF-4tosee whether they have any role in neoplastic growth of keloid.Method: We observed the expression of OPN, VEGF and ATF-4in center and marginsof keloid through HE and immunohistochemical staining. Thereafter, we observe them underthe microscope and images were acquired. Various data were obtained through variousmethods like number of positive cells classification score by comprehensiveimmunohistochemical semi-quantitative integral method of staining and comprehensive thestaining extent and the number of positive cells grading of the immunohistochemicalsemi-quantitative integral method to get the score (Count data). At the same time, imaginganalysis was done by Image-pro plus6.0software for integrated optical densitymeasurement (IOD). Respectively, statistical analysis was performed.Results:1、Comparison in expression of OPN and VEGF in central part and marginal parts of normal skin and keloid was statistically significant (P<0.05).2、 The expression of ATF-4in keloid is higher than that in normal skin and wasstatistically significant (P<0.05);the expression of ATF-4in the central part compared to themarginal part of keloid was not statistically significant (P>0.05).3、There was a positive correlation between OPN and VEGF in the central part and themarginal part of keloid (p<0.05).Conclusion:1、The expression of OPN and VEGF densely infiltrated in keloid than in normal skin,and in keloid as a whole the marginal part is densely infiltrated than the central part.2、There was an existence of positive correlation between OPN and VEGF in the centralpart and the marginal part of keliod.3、The expression of ATF-4in keloid was significantly higher than that in normalskin.4、OPN, VEGF and ATF-4can provide new ideas and theoretical basis in neoplasticgrowth of keloid.
