Quercetin whose chemical name is4H-1-Benzopyran-4-one,2-(3,4-dihydroxyphenyl)-3,5,7-trihydroxy-Flavone is a flavonol widely distributedin plants. Glycosylated forms include rutin, quercetrin,hyperin and so on.Recent research indicated, except its antioxidation and anti-inflammatoryproperties, quercetin have the effect of anti-tumor,anti-fibrosis and protectionto cardiovascular.Neurodegeneration disease is the umbrella term for the progressive loss ofstructure or function of neurons, including death of neurons. Many neurodege-nerative diseases including Alzheimer’s, Parkinson’s, and Huntington’s occuras a result of neurodegenerative processes.The aim of our researches was to study the protective effect of quercetinon the central nervous system in vivo. Firstly, memory injured mice induced bysodium nitrite was used. Morris water maze was used to study the behaviorchanging, and analyzed of the activity of SOD and MDA were used to studythe probable mechanism. Secondly,the MPTP-induced Parkinson’s diseasemodel was used. And motor behavior analysis with the rotarod test, the analyseof the activities of GPx,SOD,Na+, K+-ATPase, AchE, the immunohistochemicalof4-HNE and the measure of the level of DA were used to study the probablemechanism.Methods:1. The protective effect of quercetin on memory injured mice induced bysodium nitrite(1) Behavior tests (Morris water maze)(2) The activity of SOD and the level of MDA of the brains 2. Effect of quercetin in the MPTP-induced Parkinson’s disease model(1)Motor behavior analysis with the rotarod test(2) The measure of the activities of GPx, SOD, Na+, K+-ATPase, AChE.(3)Immunohistochemical of4-HNE(4)The measure of the level of DAResults:1. The protective effect of quercetin on memory injured mice induced bysodium nitrite(1) The result of Morris water maze showd that quercetin could reduce thethe latency time which lengthen by sodium nitrite, enhance the spatial learningability of the mice.(2) The bioassay of the brain showed that quercetin could enhance theactivity of SOD and reduce the level of MDA.2. Effect of quercetin in the MPTP-induced Parkinson’s disease model(1) The result of rotarod test showed that quercetin could enhance thebalance coordination of the mice.(2) The bioassay of the brain showed that quercetin could enhance theactivity of GPx,SOD, Na+,K+-ATPase and AchE of the brains.(3) The result of immunohistochemical of4-HNE showed that quercetincould reduce the expression of4-HNE in the brain.(4) Quercetin could increase the level of DA significant.Conclusion:1.Quercetin could enhance the activity of antioxidant,reduce the lipidperoxidation to protective the central nervous system, reduce the injure of nervecell caused by free radical, protective the ability of learning and memory ofmice.2. Quercetin could enhance the oxidation resistance of the brain byenhance the activity of GPx,SOD,Na+,K+-ATPase,reduces the level of acetylcholine by enhancing the AchE activity, which may mediate quercetininduced improvements in motor balance and coordination of MPTP-treatedmice. The enhancement of dopamine content by quercetin may restore thechanges in locomotor activity. The reduce of the expression of4-HNE showedthat quercetin reduce the lipid peroxidation and the enhancing of Na+,K+-ATPase showed that quercetin could maintain the resting membrane potentialof neurons...
