Objective:In the modern life sciences research, Gene engineering mouse life has become importantsource of human disease animal model. Gene engineering mouse life has a high application valueof the research of the occurrence, development and treatment of human diseases. Beforeapplication scientific research of animal model of human disease wich was made usingtransgenic or knockout methods, the researchers must research its biological characteristicsthoroughly. Study on the biological properties of genetic engineering animal before using is thebasic conditions, and these studies also are the key content of the current genetic engineeringanimal model.ApoE-/-Mice is an important animal model wich is used to research the pathologicalandmechanism of atherosclerosis and vulnerable plaque. Through to fed ApoE-/-mice with high fatdiet, to research AS animal model more applications. But as an animal model of atherosclerosisApoE-/-mice feeding common feed to aging time, the related biological trait and data do nothave detailed reports. these datas can infer treatment and research of human AS late onset.Thispoint is particularly important. By feeding the ApoE-/-mice fed with normal diet to18months ofage, this research observate the pathological changes of Liver and main vasculars such ascoronary artery, basilar artery, carotid artery, abdominal aorta, renal artery, and test a part ofsensitivity index of AS. The data can provides basic background information for pathologicalchanges of ApoE-/-mice fed with common feed to the vascular aging. and To provide referencefor research workers who use the ApoE-/-mouse to make study.Methods:1、Selected ApoE-/-mice male of February age of8, the same age and gender C57BL/6J miceof8served as the control group, raised in the experimental animal center of Shanxi MedicalUniversity barrier environment facilities. Feed (fed by high temperature and high pressuresterilization) to18months of age, temperature20-23℃,humidity45-55%,light executes12hoursof turnover. Select the main artery and liver were stained with HE,analyze and deal with the HEstaining results with image analysis system.2、Extract animal abdominal aortic blood,and use automatic biochemical analyzer to detect theplasma biochemical indexes:total cholesterol(TC),Low density lipoproteincholesterol(LDL-C),High density lipoprotein cholesterol(HDL-C), Triglyceride(TG). 3、Using the enzyme mark instrument determination:the content of serum high sensitive Creactive protein(hs-CRP)and relative expression of Hepatic peroxisome proliferator activatedreceptor(PPAR).4、All statistical data is analyzed by Statistical Package for the Social Science13.0(SPSS13.0),all data in representation of±s, groups were compared using a single factor analysis ofvariance.Results:1、Two groups of mice fed with common feed to18months of age, there was no death.ApoE-/-mice intake decreases relatively, the coat was in amuck. Some C57BL/6J mouse appeared theemergence of hair symptom. Two groups of animal blood and liver by HE staining, lightmicroscopic observation showed that coronary artery and basilar artery of ApoE-/-mice do notform Atherosclerotic plaque. Carotid artery, abdominal aorta and renal artery form Apparent ASplaque, and Liver tissue visible part of lipid deposition.C57BL/6J mice in different parts ofvascular and hepatic pathology showed normal.2、Blood lipid testing results:TC(2.03±0.25vs13.82±2.73mmol/l P<0.05)、LDL-C(0.20±0.08vs1.88±0.76mmol/l P<0.05),HDL-C(1.52±0.13vs0.62±0.20mmol/lP<0.05),The difference was statistically significant. The difference of serum TG wasstatistically significant(2.02±0.34vs1.58±0.68mmol/l P>0.05).3、The difference of the content of serum hs-CRP was statistically significant(5.61±0.33vs6.80±0.38ug/l P<0.01).Relative expression of liver PPAR was statistically significant(247.62±21.74vs339.43±30.07ng/l P<0.01).Conclusions:1、Two groups of mice fed with common feed to18months of age, there was no death. ApoE-/-mice intake decreases relatively, the coat was in amuck. Some C57BL/6J mouse appeared theemergence of hair symptom maybe it had relations with Line defect.2、The coronary artery and basilar artery of ApoE-/-mice do not form Atherosclerotic plaque. Itshow that the site is not suitable for the study of atherosclerosis.3、The carotid artery, abdominal aorta and renal artery of ApoE-/-mice form apparent AS plaque. Itsuggested ApoE-/-mice can be used as the ideal atherosclerosis model.4、The levels of ApoE-/-mouse TC and LDL-C were significantly higher than those in normalC57BL/6J mice.TG level also had a rising trend. ApoE-/-mouse HDL-C has decreased only fornormal control rats1/4.ApoE-/-blood serum content of hs-CRP was higher than C57BL/6Jmice,and liver PRAR was higher thanC57BL/6J mice. These traits coincide with AS symptoms,and it was applicable to the pathogenesis of early detection of AS.
