Objective To investigate the inierventional effects of ketamine and midazolam.Methods Sixty Male Wistar rats were randomly divided into sham operation group (SHAM) group, cecal ligation and puncture (CLP) group and ketamine (KT) group。Each group were randomly divided into two subgroups (n=10), S1、S2、C1、C2、K1、K2group, the S1、C1、 K1group is used to the collection of specimens, the S2、C2、K2group is used to observe the survival time. The S1group and the S2group received without any treatment, other groups underwent the rat sepsis model which was established by using cecal ligation and puncture(CLP). In ketamine group, the rats received intraperitoneal injection of40mg/kg ketamine20minutes before shock.①Before the operation of CLP, The carotid artery catheterization was cannulated to monitor mean arterial pressure (MAP); The rats were inserted into the trachea tube for clean-up respiratory secretions.After the operation, give sodium chloride10ml/kg·h till executed. After the technique finishes stable15min, starts to time. And the Level of HSP70was measured by Immune-ohistochemical method. the S1、C1、K1group were sacrificed12h in rat brain tissues using immun ohistochemical semi quantitative method to detect the expression of heat shock protein70.②The S2、C2、K2group were observed, the survival time and survival rate of7d.Results (1) General situation:Compared with the Sham operation group, CLP energy group of rats presents the energetic dispirited, the activity to reduce gradually with the lapse of time, sometimes will appear is addicted to rests, trembles, will even produce the polypnoea.After the experiment is finished to death rats, the belt irritates the nose the odor, obviously the abdominal cavity bloody pus fluid seeped out, the caecum swelling nigrescence, somewhat presented the gangrene, the adhesion in the mesentery, hepatorenal and so on important organ also had the silt spot, the ecchymosis and hyperemia drops; The KT group reduces obviously.(2) The change of mean arterial blood pressure(MAP):after the model of CLP, The mean arterial pressure of all groups decrease gradually, the MAP of CLP group and KT group decrease signifieantly(P<0.01) after9h; the MAP of CLP group achieve shock instandards; the levels of MAP in the KT group were signifieantly higher than that in CLP group(P<0.01), and no shock. the hemodynamic responses were reversed in ketamine treated animals.(3)HSP70:HSP70were observe in all groups. However, positive expression of HSP70in CLP group was lower than Sham operation group, while higher than ketamin group(both P<0.01).(4)The survival rate: The survival rate was lower in CLP group (0) compared with Sham operation group (100%) and ketamine group (40%), the differences were significant (both P<0.05).5) The expression of HSP70in correlation with survival:Strengthens along with the HSP70expression, the Wistar rats survival percentage also correspondingly enhanced, and assumes is remarkable is being related (P<0.05).Conclusion The Wistar rats receives the infectious shock the stimulation to cause in the brain organization seahorse area the HSP70expression to increase, and along with the hot shock protein70expression’s enhancements, the Wistar rats’s survival time also correspondingly lengthens, explained that the ketamine possibly through induces increases in the brain organization the hot shock protein70expressions to have the protective effect to the Wistar rats, after this experiment confirmed that may think the ketamine and the brain protective function present correlational dependence.
