Objective:To investigate the expression and clinical significance of bone morphogenetic protein 2(BMP-2)in bone tissue around the implant after simvastatin intervention.Method:After 1-week adaptive feeding without abnormal condition,40 female adult SD rats were equallydivided into A group(normal control group)and B group(experimental group group). A groupwas intraperitoneal injected with citrate buffer. B group was intraperitoneal injected withstreptozotocin(STZ)50mg/kg to set up the diabetic rats model. After the model wassuccessfully established,A group and B group were implanted with Titanium in the proximaltibial metaphysic.B group was randomly allocated to B1group(Diabetes Mellitus+ implantgroup)and B2group(Diabetes Mellitus+ implant + simvastatin group). There were 10 rats ineach group. B2 rats were given orally simvastatin 5mg·(kg-1·d-);A and B1 rats were givenorally normal saline 1.5ml·(d-1). After of 12 weeks of feeding the rats were sacrificed.Immunohistochemical technique were used to assess the expression of BMP-2 in bone tissuearound the implant. Results:Positive rate of cells in B2 group was significantly higher than that in B1 group. The differenceis significant statistically(P < 0.01).Conclusion:Simvastatin can enhance the expression of BMP-2 gene in bone tissue, improve bonemetabolism and implant osseointegration. |
