Objective To investigate the expression tend of ICAM-1protein in different time afterbrain contusion by immunohistochemical SABC and western blot, establish the relation betweenexpression of ICAM -1 after brain contusion and early-injury time. The sequential expression ofICAM-1 was used for the estimation of early posttraumatic intervals after cerebral contusion inforensic practice.Methods the fifty-six SD rats are choose to the control group and experimental group, eachguop is eight rats, the rat model of cerebral contusion was established. the samples taken at1h,24h,48h,72h,7d after cerebral contusion. The control group of rats are immediate killed .Therat brains are fixed with 4% Para formaldehyde by Immunohistochemical and HE, and othersamples are stored in -80℃liquid nitrogen for the isolation of protein.Results 1) The result of HE: the morphology of neuronal cell is normal incontrol group, pale is nuclear. Brain parenchyma of experimental group wasbleeding, the injury brain can be seen localized subarachnoid hemorrhage. Neuronalcell edema, cytoplasmic becomes empty, the nuclear pale,gradually developed intoa nuclear fragmentation dissolving, neuronal was necrosis.2) The result ofimmunohistochemisty:1h after cerebral contusion can observe positive cells scattered insmall amount, positive product was brown yellow, mainly located in the vascular endothelialcells and glial cells. The expression of ICAM -1 protein observed at 24h was increased, peakedat 72h.the evascular endothelial cells and glial cells are widely expressed yellow product, andsome nucleus in neurons also appear positive cells. The expression of ICAM-1 in theexperimental group on the 7th day after injury still was significantly higher than that in thecontrol group. 3) The result of the Western Bloting Image analysis technique is used toImmunoblotting of strip integral optical density, The expression of ICAM -1 proteinwas observed at 1h after injury ,increased at 24h,reached the peak at 72h and thenwas decreased. The expression of ICAM-1 in the experimental group on the 7th dayafter injury still was significantly higher than that in the control group .Conclusions: The expression of ICAM -1 after cerebral contusion was regularlychange with the time going. Immunohistochemical and Western Bloting are used toevaluate the expression of ICAM -1 after cerebral contusion in rats. This sequential expression of ICAM-1 is used for the estimation of early posttraumaticintervals after cerebral contusion in forensic practice.
