| Objective:To investigate the effects of adiponectin on oxidative stress and apoptosis in Retinal pigment epithelium cultured with high glucose.Methods:The cells were divided into three groups:the normal glucose group, high glucose group and high glucose plus adiponectin group. After culture for24h,48h,72h Oxidative stress was evaluated by detecting levels of MDA and SOD in the supernatant of culture media. The expression of p66Shc, Ho-1was detected by Real-Time PCR. Flow cytometry were designed to observe and detect cellular apoptosis.Results:Our findings showed significant increase of MDA levels and decrease of SOD activity in the high glucose group compared with the control group. However in the adiponectin group MDA levels decreased and SOD activity increased significantly compared with those in the high-glucose group. Compared with the control group, high glucose increased Ho-1mRNA expression; adiponectin group also expressed a high glucose group increased. Compared with the control group, high glucose p66Shc mRNA expression increased; the adiponectin group also expressed a high glucose group decreased, showed a time-dependent. The apoptotic rate of Retinal pigment epithelium in the high glucose group increased significantly compared with that in the control group while the apoptotic rate in the adiponectin group declined remarkably in comparison with that in the high glucose group.Conclusion:The adiponectin can rectify high glucose-induced apoptosis of RPE, play a protective effect on Retinal pigment epithelium by upregulating the Ho-1expression and downregulating p66Shc expression. |
