| The apoptosis of RPE cells played an important role in retinal degeneration diseases. But the mechanism of apoptosis is very complicated, so it's necessary to identify the mechanism of apoptosis of RPE cells. The important role of mitochondria-mediated apoptosis has been proved. This research aimed at the role of AIF-mediated way in the apoptotic process of RPE cells, which is independent on Caspase. ObjectiveTo study the expression of AIF in the apoptotic process of RPE cells, especially the translocation from mitochondria to cytoplasm of AIF. MLT is used to test the role of AIF in the apoptosis of RPE cells in profiles.MethodsThe apoptotic model of cultured RPE cells was build up with H2O2. Then the display of ROS, apoptotic ratio of RPE cells, mitochondrial membrane potential and the proliferative ratio were detected. Western blot, real-time fluorescent quantitative polymerase chain raction and immunofluorescence were used to detect the translocation of AIF. Then MLT were added to detect the impacts on AIF's translocation. All of the changes were analyzed to enrich the mechanism of apoptosis of RPE cells.ResultsIn the model, mitochondrial ROS increased with the increasing concentrations and durations of H2O2. The change of mitochondrial membrane potential was not conspicuous. The expression of AIF in cytoplasm increased. MLT can decrease the production of ROS and the apoptotic ratio of RPE cells. But MLT increased the expression of AIF both in cytoplasm and mitochondria.ConclusionAIF takes part in the apoptotic process of RPE cells. But the status of this factor is not very clear, which needs additional research to identify. On the other hand AIF may plays a secondary role in the apoptotic process of RPE cells through the interference of MLT. |
