AIMTo investigate the killing effect of TK/GCV system combined with doxorubicin on humanteosarcoma cells and study the efficiency of killing the human osarcoma cells by TK/GCV system combined with doxorubicin.Methordsplasmid pIRES-EGFP-tk.and transfect osteosarcoma cell line mg-63, cell circle analysis and under fluorescence microscopy observe cell growth features,then add the GCV and doxorubicin,study the efficiency of killing the osteosarcoma cells by TK/GCV system combined with doxorubicin.Results1. Successful construction restructuring plasmid pIRES-EGFP-tk, The results showed that TK gene was integrated into mg-63cells.2.Observed under fluorescence microscope mg-63(TK+) showing green fluorescence.3.Flow cytometric detection ZhuanRan rates in61.4.The growth of mg-63(tk+) cells was greatly inhibited by GCV administration, with1mg/l as lowest concentration. Doxorubicin killed mg-63cells at concentration of1mg/l. When combined with low concentration of GCV5μg/ml the doxorubicin can be decreased to0.25μg/ml.
