Objective:In recent years, the incidence of breast cancer is increasing year by year,and has leapt to the forefront of the female various malignant tumor. For the lack of high sensitive and minimally invasive diagnosis, there are still difficulties in early diagnosis of breast cancer. This study was designed to search for differentially expressed proteins in health women and breast cancer patients by using SELDI-TOF MS, aims to screening meaningful protein which used for early diagnosis of breast cancer and breast cancer with the transfer of the axillary lymph node.Methods:Serum samples collected from84breast cancer patients and50health women.84breast cancer patients include39breast cancer patients with axillary lymph node metastases and45breast cancer patients without axillary lymph node metastases. All samples were detected by surface-enhanced laser desorption/ionization time-of-flight mass spectrometry (SELDI-TOF-MS) at the PH6.0Buffer with serum was diluted40times experimental conditions. Differences of mass spectrums between health women and breast cancer patients with axillary lymph node metastases and without axillary lymph node metastases by Biomarker Wizard software. The data were statistically analyzed by SPSS17.0software, finally evaluate the meaning proteins diagnostic efficiency.Results:The data of breast cancer patients and health women were compared and13discrepant protein peaks were found (P<0.01). Area under the curve of ROC of Protein peaks (M/Z)4206is0.915. The protein peaks of m/z4206Da have the highest value diagnostic with a sensitivity of92.85%and specificity of94%.But there was no statistically meaning in different Clinical stages. The protein peak of m/z4781Da has significant expression in group with axillary lymph node metastases. Have the highest value diagnostic with a sensitivity of84.62%and specificity of88.89%. In validation test, the sensitivity, specificity and correct ratio of the method were85.71%,90.63%and88.33%respectively. But there was no relationship with the involvement of lymph node numbers.Conclusion:SELDI-TOF-MS technology can be used to screen serum markers of breast cancer patients and disease process judgment. The protein (M/Z4206) may be as a new tumor marker markers in diagnostic of the breast cancer and the protein (M/Z4781) may be as a new tumor marker in prediction of the breast cancer with axillary lymph node metastases.
