Objective:To study the changes of T lymphocyte subsets and T regulate cells in peripheral blood in children with infection mononucleosis(IM), and to explore the role of CD4+CD25+CD127-T regulate cells in the pathogenesis of IM. Meanwhile, to explore whether there is any correlation between lymphocyte subsets and EBV load.Method:1.Experimental object:47children with IM and EBV-DNA load> 5.0*102copies/ul for observation group,32 healthy children in the same period as control group subjects. Each one was drawn lml and 2ml peripheral blood, anticoagulated.2.Index detection A flow cytometry: a plus antibody:Test tube is appended with CD4-PC5、CD25-FITC、CD127-PE monoclonal antibody and CD4-FITC/CD8-FE/CD3-PC5 three mark monoclonal antibodies each 5ul, then each test tube appended with 20ul anticoagulate complete blood, mixed, away from light 20 minutes.b Hemolysis:each test tube is appended with 90ul hemolysin, mixed, ecaded from light 15minutes, then centrifuged for 5 minutes, by 1200r/min, deserting the upper liquid, awaiting to be detected. We choose 10000 cells from each sample, and detecting the expression of CD3+、CD4+、CD8+、CD4+CD25+andCD4+CD25+CD 127-by flow cytometry CELLQUGST. B the detection of EBV-DNA load:each test tube was drawn 2ml peripheral blood at the state of hollow by the ABI-7300-FQ-PCR. Detecting steps、judgment for the results and the quality control are detection limit is 5*102copies/ul, one which is equal to or greater than this value is positive.Results:Compared with the following results in controls,CD3+,CD3+CD8+were increased and CD3+CD4+,CD4+/CD8+ratio,CD4+CD25+,CD4+CD25+CD127-were reduced in the IM patients at the acute stage. There is a positive correlation between EBV loaf andCD3+,CD8+ during acute phase. there is a negative correlation between EBV load and CD4+ and CD4+/CD8+ ratio, regulation T cells during acute phase.Conclusion:There lies the immune dysfunctions in the children with acute infectious mononucleosis. the degree of immune dysfunction is relevant to EBV-DNA copy number. meanwhile, regulation T cells of peripheral blood in the IM patients was diminished, they possible play an important role in the pathogenesis of IM.
