Objective To investigate the effect of pantoea agglomerans lipopolysacc-haride(LPSp) combined with hepatitis B surface antigen(HBsAg) on IL-4, IL-6 IFN-y and TLR4 expression in the pregnant mice.Methods Fifty pregnant mice were randomly divided into five groups: control group, HBsAg group, combined with low dose group, combined with middle dose group, combined with high dose group,10 pregnant mice in each group. When the first day and the fourteenth day, pregnant mice of the five groups were respectively injected into the muscle with 0.9% NaCl respectively, HBsAg 100ul, HBsAg 100ul+LPSp 5ug, HBsAg 100ul+LPSp 15ug, HBsAg 100 ul+LPSp25 ug, collected the serum and placenta of the pregnant mice when the twentieth day, then the IL-4、IL-6 and IFN-y were detected by ELISA, the protein and mRNA expressions of TLR4 in the pregnant mice were detected by immunohistochemistry and reverse transcription-polymerase chain reaction (RT-PCR). Measured the length, weight and brain weight of the neonates.Results (1)The IFN-y levels of each conbined group were significantly higher than control group and HBsAg group, in concentration-dependent manner, (P<0.01), the IL-4 and IL-6 levels of each conbined group were significantly higher than control group (P<0.01), but there were not significant differences between the combined with high dose group and HBsAg group(P>0.05), the IFN-y levels of the HBsAg group were significantly lower than control group, the IL-4 and IL-6 levels of the HBsAg group were higher than control group(P<0.01). (2) LPSp increased TLR4 protein expressions in placenta of the pregnant mice, the protein expressions of TLR.4 in each conbined group were higher than control group and HBsAg group (P<0.01).(3) LPSp increased TLR4mRNA expressions in placenta of the pregnant mice, the expressions of TLR4mRNA in each conbined group were higher than control group and HBsAg group (P<0.01). (4) There was no difference in length,weight and brain weight in different groups (p>0.05).Conclusions (1) There was no side-effects such as intrauterine growth retardation occurred in the neonates.(2) LPSp dose-dependently modulated the Thl/Th2 cytokines, high dose of LPSp increased the levels of Thl cytokines, low dose of LPSp increased the levels of Th2 cytokines.(3) LPSp increased TLR4 protein and TLR4mRNA expressions in placenta of the pregnant mice, and LPSp maybe not cause placental morphologic change.
