Objective:To study the IL-4, IFN-y and TLR4expression induced by different density of LPSp in the cord blood mononuclear cells of HBV intrauterine infection in vitro and research the mechanism of LPSp and HBV intrauterine infection.Methods:Selected71pregnant women with HBsAg, HBeAg positive, normal liver function of peripheral blood. Cord blood of HBV-DNA positive for the intrauterine infection group, a total of25cases of cord blood. HBV-DNA negative for intrauterine uninfected group, a total of46cases. Collect cord blood during delivery, heparin, isolated using Ficoll density centrifugation CBMC and incubated for24hours. According to different inducing concentration of LPSp, the Intrauterine infection group is divided into4sub-group:no-LPSp induced group, LPSp1μg/L induced group, LPSp10μg/L induced group, LPSp100μg/L induced group. The control group include20normal pregnant women with HBV markers negative. Separated and cultured the CBMC. Using a Enzyme Hnked immunosorbent assay (ELISA) in culture supernatants of IFN-y, IL-4levels, measured by flow cytometry CBMC surface expression of TLR4.Results:(1) In intrauterine infection group, the level of IFN-γ in CBMC cell culture supernatant was lower than the intrauterine uninfection group and control group (P<0.01), IL-4levels higher than the intrauterine uninfected group and control group (P<0.01). Intrauterine infection group and the control group compared to IFN-γ, IL-4level was no significant difference (P>0.05).(2) In intrauterine infection group, The IFN-y levels of each LPSp group are higher than no-LPSp group(P<0.05), the result of IL-4levels contrary to IFN-y(P<0.05).(3) Each group of IFN-y levels LPSp concentration was positively correlated (r=0.778), levels of IL-4LPSp concentration was related (r=-0.927).(4) Each group of TLR4expression was positively correlated with IFN-y levels(P<0.05). TLR4and IL-4was negatively correlated(P<0.05). TLR4expression level and LPSp concentration was positively correlated (r=0.754).Conclusion:CBMC of TLR4is LPSp to cause the immune response of the receptor. LPSp can induce up regulation of CBMC of TLR4. LPSp can adjust the CBMC secretion of Thl/Th2cytokines in a dose-dependent. High the dose LPSp enable increased secretion of IFN-y, and low the dose LPSp can make to an increase in IL-4secretion. Therefore, different concentrations LPSp may affect TLR4signaling pathway induced CBMC secrete different cytokines, selective regulation of Thl/Th2responses.
