ObjectiveTo investigate the clinical significance and mechanism of WWOX gene abnormal expression in acute leukemia the same as FHIT gene and p73gene.Methods107cases bone marrows from acute leukemia patients and31cases bone marrows that weren’t from leukemia paients (as control) were collected. The level of WWOX、FHIT and p73transcripts in acute leukemia patients and control cases were assayed using reverse transcriptase-polymerase chain reaction (RT-PCR), Methylation PCR (MSP) were used to detect the promoter methylation of WWOX gene、FHIT gene and the first exon methylation of p73gene. To analyse the expression of the three genes mRNA and the methylation status of the same three genes, also the correlation between the genes mRNA expression and the methylation status of theirs.Results1.The rate of WWOX mRNA positive expression was47.66%in acute leukemia, which was significantly different from that of control group as93.55%; the promoter methylation rate of WWOX gene was42.06%in acute leukemia, which was also significantly different from control group. There was negative correlation between the WWOX mRNA positive expression and the promoter methylation status in acute leukemia, and the correlation coefficient was-0.661.2. The rate of FHIT mRNA positive expression was44.86%in acute leukemia, which was significantly different from that of control group as96.77%; the promoter methylation rate of FHIT gene was43.93%in acute leukemia, which was also significantly different from control group. There was negative correlation between the FHIT mRNA positive expression and the promoter methylation status in acute leukemia, and the correlation coefficient was-0.685.3. The rate of p73mRNA positive expression was53.27%in acute leukemia, which was significantly different from that of control group as93.55%; the first exon methylation rate of p73gene is36.45%in acute leukemia, which was also significantly different from control group. There was negative correlation between the p73mRNA positive expression and the first exon methylation status in acute leukemia, and the correlation coefficient was-0.536.4.There were positive correlation between WWOX gene mRNA expression and FHIT gene mRNA expression, the correlation coefficient was0.569, while there were no correlation between the promoter methylation of WWOX gene and the promoter methylation of the FHIT gene; There were positive correlation between WWOX gene mRNA expression and p73gene mRNA expression, the correlation coefficient was0.556, while there were no correlation between the promoter methylation of WWOX gene and the first exon methylation of the p73gene. Conclusions:1. The expression of WWOX、FHIT and p73genes in acute leukemia patients were usually absent or reduced, they may take an important effect in the pathogenesis and development of acute leukemia.2.The WWOX、FHIT and p73genes were abnormal methylation in acute leukemia, they may be the dangerous factor during the process of occurrence and development in the acute leukemia.3.It was found that the expression of WWOX、FHIT and p73genes were negative correlation with the methylation status of their genes expectively, which suggested the gene abnomal methylation may be result to gene silence, and the transcripts reduced or absent.4. The expression of WWOX was positive correlation with the expression of FHIT in acute leukemia, while there were no correlation between the methylation status of WWOX gene and the FHIT gene, which was also the same between WWOX gene and p73gene.
