Objective:To investigagte the mutation status of k-ras gene in colorectal cancer and analyse the associations between its mutation status and clinicopathological characteristics in colorectal cancers so as to provide better service for clinical treamtment.Method:The specimens of34patients with colorectal cancer were collected,DNA sequencing were performed in these tissues to tetect k-ras gene mutations at codon12th and codon13th of exonl,and simultaneously collect these patients clinicopathological and treatment data.Result:Of total34cases,12cases of k-ras gene mutations were detected,the mutation rate is35.3%,among these12cases,9cases at codon12and3cases at codon13.There were no significant differences in gender, age, primary tumor site, differentiation, depth of invasion, lymph node metastatic, TNM stage and distant metastasis (all P>0.05).There is no significant difference between the treamtment efficacy and adverse events when colorectal cancer patients use FOLFOX as chemotherapy. Conclusion:1.The k-ras gene mutation rate of colorectal cancer is34.3%, mutations position mainly at codon12,secondly at codon13.2. There were no significant differences in gender, age, primary tumor site, differentiation, depth of invasion, lymph node metastatic, TNM stage and distant metastasis.3. K-ras gene mutation is not related to the efficacy of FOLFOX chemotherapy, can not be used as predictive factor of efficacy of chemotherapy.4. K-ras gene mutation probably can not be used as predictive factor of which the occurrence of adverse reaction and its severity of chemotherapy in patients with colorectal cancer.
