Objective:To investigate the expression and clinical significance of NDRG1 (N-myc downstream regulated gene-1) in the carcinogensis and progression of cervical cancer. Also to study the express of P16 in cervical cancer, and the correlation with NDRG1, for investing the mechanisms of both genes expression.Method:NDRG1 and P16 protein expression were detected by Immunohistochemistry in 30 cases of normal cervical tissue,120 cases of cervical cancer, the relationships between the expression and cervical cancer were analyzed.Results:The positive rate of NDRG1 expression in normal cervical tissue was 70.0%, while in cervical cancer tissue was 22.5%. From normal cervical to cervical cancer tisuuse, NDRG1 progein was gradualy decreased (χ2=24.885, P<0.05).The results of IHC shows:There was no obvious correlation between NDRG1 positive expression with differentiation degree, pathologic type and clinical stages. In cervical cancer group with lymphnode metastasis, the stronge positive (+++) raito was zero (0/16), most of them are weak positive. In cervical cancer group with non-lymphnode metastasis, the strong positive (+++) raito was 24.6%(16/65), most of them are strong positive. For cervical cancer cases, there was inverse correlation of NDRG1 strong positive expression and metastatic conditions (r=-0.235,P<0.05). The positive rate of P16 expression in normal cervical tissue was 26.7%, while in cervical cancer tissue was 82.5%. From normal cervical to cervical cancer tisuuse, P16 protein was gradualy increased (χ2=36.587,P<0.05). For cervical cancer cases, there was no obvious correlation between P16 positive expression with differentiation degree, pathologic type and clinical stages. In cervical cancer group with lymphnode metastasis, the positive raito of P16 was 100% (16/16); In cervical cancer group with non-lymphnode metastasis, the positive raito of P16 was 78.5%. There was positive correlation of P16 protein positive expression and metastatic conditions (r=0.227,P<0.05). High P16 protein positive expression indicated high lymphnode metastasis ratio. The expression of NDRG1 was inverse correlation with P16(r=-0.277,P<0.05).Conclusion:The low expression of NDRG1 and the obvious high expression of P16 protein in cervical cancer, showed both were related with the occurrence of cervical cancer. NDRG1 gene was down-regulated in cervical cancer, suggesting that NDRG1 maybe a potential biomarker for predicting cervical cancer.
