Objective:Paclitaxel plus platinum is the most commonly chemotherapy regimen of ovarian cancer, now known mainly through its direct cytotoxicity to kill tumor cells, but whether its indirect involvement in immune regulation is not known. CD4+CD25+Treg cells affects the immune system response to tumor antigens, thus affecting the generation and development of tumor. The FOXP3transcription factors express in CD4+CD25+Treg cells and influence its development and the function, so, its can be seen as a specific marker. This study collects60cases with advanced ovarian epithelial carcinoma. We collect their serum samples at different times and use flow cytometry to test positive rate of CD4+CD25+FOXP3+cells. And we will observe the relationship between the changes and the efficacy of chemotherapy. And to observe whether it can be used as a effect forecast index for advanced epithelial ovarian cancer before chemotherapy.Methods:Case selection:A total of36cases in the group. All the cases were histologically confirmed as epithelial ovarian cancer, postoperative relapse or palliative debulking surgery, ECOG scores≤2. Outcome measures:①.Before chemotherapy, general information of patients, tumor size and peripheral blood CA125value should be recorded. FACS detects the positive rate of peripheral blood CD4+CD25+FOXP3+cells. Simultaneouly, we detect the ratio of CD4+/CD8+cells by FACS.②. We record the general data of patients and tumor size, peripheral blood CA125value in different cycles of chemotherapy. We evalued the efficacy of chemotherapy. And we use FACS to detect the positive rate of peripheral blood CD4+CD25+FOXP3+cells and the ratio of CD4+/CD8+cells after different cycles of chemotherapy.③. Before and after treatment, we record blood routine test and biochemical series check. We make follow-up records by B ultrasound, CT and MR. And we record toxicity, quality of life and treatment effect.④. The endpoint of observation was the progress of disease and we record the time to progresslon.Results:The positive rate of fCD4+CD25+FOXP3+cells decreased significantly in the patients with CR+PR,especially in patients with PR.While in the progressed after chemotherapy, there is no obvious changes, and it even has a tendency to rise. CD4+/CD8+ratio in the sensitivity to chemotherapy (CR+PR)elevated,and in the progression,it has no change,or even declines.Conclusion:The changes of CD4+CD25+FOXP3+cells positive rate in the pa.tients with advanced epithelial ovarian cancer before and after chemotherapy related to the efficacy of chemotherapy,which can be used as a predictor of chemotherapeutic efficacy...
