Single-walled carbon nanotubes (SWNT) is a kind of nano-materials with regular structure, stable chemical property. As the ability to enter cell by endocytosis, SWNT with suitable size is potential drug carriers. SWNT has obvious advantages because of its targeting of tumor tissue. siRNA can effectively silence the gene expression of tumor over and interrupt the abnormal development of tumor signaling, in order to achieve tumor suppression, is the current hot spots in cancer therapy.This study intends to connect siRNA with SWNT and develops SWNT as a siRNA delivery. First, in order to reduce the aggregation, SWNT was functioned with DSPE-PEG-Amine by ultrasonic treatment. Second, siRNA was connected to SWNT by the different bifunctional crosslinker sulfo-LC-SPDP. Then, the transfection efficiency of SWNTs-siRNA to tumor cells was detected by flow cytometry. Finally, siRNA-SURVIVIN、 siRNA-MDM2and siRNA-β-CATENIN were connected to SWNT, and were acted on breast cancer cells. Cell proliferation was delected by MTT assay and apoptosis was delected by Annexin V/PI kit.The concentration of functioned SWNT was delected as50mg/L by Near infrared spectroscopy. siRNA was connected with SWNT with the efficiency of83.55%. A good transfection efficiency was delected when the concentration of siRNA connected wih SWNT was above60nM.82.43%of transfection efficiency was delected with1OOnM of siRNA used. SWNT carriers showed no obvious cytotoxicity, and the inhibition of cell proliferation rate was below10%. Treated with three kinds of SWNT-siRNA, the proliferation of B-Cap-37cells was inhibited, and the apoptosis was promoted. The best proliferation-inhibiting was achieved with SWNT-siRNA-MDM2. The most apoptosis was achieved with SWNT-siRNA-SURVIVIN.From this study, siRNA was connected with SWNT with a great efficiency. And great transfection efficiencies were achieved when The SWNT-siRNA of above60nM intransfected B-Cap-37. Three kinds of SWNT-siRNA can effectively suppress the proliferation of breast cancer cells and promote apoptosis.
