It is very common and effective in clinic that doctors use Hemodialysis totreat high cholesterol patients. Because of its special molecular structure and spaceconformation, heparin has good blood compatibility and specific reaction with LDL. Soheparin modified resin to be the LDL adsorbent is one of the hottest researches.However, a lot of clinical data show that, the adsorbent’s poor blood compatibility andselectivity to LDL cause many side effects to patients. In this paper, based on improvingadsorbent’s adsorption rate and selectivity to LDL, and adsorbent’s blood compatibility.We prepared a heparinized resin adsorbent for clearing LDL in blood plasma. Thisresearch can provide a theoretical basis for explore blood purification material hasbetter treatment.About our study:Objective:Heparin was covalently immobilized onto the Polystyrene macroporous resinsurface to prepare a blood purification material with high blood compatibility andspecific absorbability to LDL.Methods:FT-IR was used to demonstrate the success of covalently immobilizing heparinonto the resin’s surface; Toluidine blue spectrophotometric was used to analyze heparindensity on resin’s surface; Coagulation times were measured to represent modifiedresin’s blood compatibility;LDL solution,HSA and LDL mixed solution,serumadsorb test were experimented to test its absorbability.Result and conclusionResults:(1) Heparin was successfully immobilized onto the surface of XAD1600resin bycovalent bond; and the density of heparin immobilized on1g XAD1600resin was about68.5μg; Modified resin’s Coagulation times PT and TT were longer than XAD1600resin within normal limits.(2) Adsorption LDL form LDL solution(PH=7.4), adsorptive ratio(adsorbent/g:protein solution/=1:5) for LDL was82.6±2%; LDL,HSA solution,adsorptive ratio (adsorbent/g:protein solution/=1:5) for LDL was81.3±2%, butadsorption ratio for HSA was7±2%; Adsorption LDL,HSA,Globulin from serum, Adsorptive ratio (adsorbent/g:protein solution/=1:5) for LDL was82.4±2%,the otherswere all less than10%.Conclusion:High selective adsorption for LDL and good blood compatibility hadbeen presented in medical adsorbent for blood purification, so this search could providerationale for clinical study.
