Objective:To investigate the effects of triptolide on the proliferation and apoptosis induction in epidermal squamous cell carcinoma cell line A431 in vitro.Methods:Human epidermal squamous cell carcinoma cell line A431 was cultured. After the treatment with triptolide, inhibition of cellular growth was determined by measuring MTT dye absorption of the living cells. Light microscopy observed morphological changes. The DNA content and apoptosis were assessed by flow cytometry.Results:Triptolide could significantly inhibit the proliferation of A431 cells with a dose- and time-dependent manner. Light microscopy revealed characteristic morphological changes of cell apoptosis. The DNA flow cytometric analysis showed that triptolide could induce more sub-G1 phase in A431 cells, and cell cycle progression analysis which performed by flow cytometry indicated that cell cycle was obviously inhibited by triptolide in G0/G1 phase.Conclusion:Triptolide plays an inhibitory role in the tumor cell proliferation and induces the cell apoptosis in epidermal squamous cell carcinoma cell line A431.
