ObjectiveTo investigate the effect of Ruan Mai Ling on the migrationinduced by platelet derived growth factor(PDGF) in vascular smoothmuscle cells(VSMCs) from rats.Methods1.Cultured VSMCs derived from rat thoracic aorta were used.2.VSMCs were identified by smooth muscle actin monoclonalantibody.3. Ruan Mai Ling drug serum was prepared by from rats after5days drug garage.4.The effect of Ruan Mai Ling on F-actin polymerization wasdetected by FITC-phalloidine labeling and examined by confocalmicroscope;5.Cell migration was determined by modified Boyden chamberassays;6.Atovastatin was used as a positive control.Results1. PDGF stimulated a increase in stress fiber and pseudopodsformation. Atorvastatin reduced pseudopods and tension fiber. Pseudopods and tension fiber were decreased in the presence of10%Ruan Mai Ling drug serum.2.Ruan Mai Ling drug serum produced an inhibition of themigration induced by PDGF-BB,the peak inhibition rate was51.7%(P<0.01).ConclusionsRuan Mai Ling significantly inhibited the migration of VSMCsand cell skeletal reorganization induced by PDGF.
