OBJECTIVE: To establish a rat model of gestational diabetes (GDM) by injectedstreptozotocin (STZ) unilateral intraperitoneally. Observe the impact of GDM on the structure ofthe fetal heart and master the T-box5transcriptional factor (TBX-5) expression in the heart offetal rats. Thus elaborated the role of the TBX-5induced heart developmental defect in the fetalrats of GDM.METHODS: The Sprague-Dawley female rats were randomly divided into two groups. Aftermating success, the female rats in GDM group were injected with STZ and the female rats incontrol group were injected with PBS in the same position respectively. Seventy two hours afteradministration, both groups were began to detecte body weight and blood glucose regularly. Twogroups of pregnant rats were randomly selected after cesarean12,15,19days, all rats wereuterine-incision delivery and got the embryo heart tissues,then observing the pathologicaldiagnosis of heart tissues by HE stain, the distribution and expression of TBX-5in heart tissueswere analyzed by immunohistochemical techniques.RESLUT:1. The general growth of female rats: since pregnant3days, GDM group female rats began toincrease in drinking and eating, increase urine output, hairs chaos, gloss variation, while thecontrol group female rats had no significant changes. Compared with control group, maternalbody weight of3,6days gestation in GDM group was no significant difference, P>0.05. Bothgroups weight showed an increase, however, after9days gestation, the GDM group were lowerthan the control group,the difference was statistically significant, P <0.01.Compared with thebeginning of the modeling (3.81±0.70mmol/L), blood glucose at three days of pregnancy inGDM group (15.00±0.76mmol/L) was significantly increased,,the difference was statisticallysignificant, P <0.01. Since3days of gestation, GDM group blood glucose higher than the controlgroup at different time points,,the difference was statistically significant, P <0.01. The controlgroup no significant change in blood glucose in the whole pregnant days, the difference was notstatistically significant, P>0.05. The mother rats of GDM group abortion in pregnant12days significantly increased compare to the control group, with statistical difference, P <0.05. Inpregnant15,19days the number of abortions was not statistically significant, P>0.05.2. The general development of embryos: Compare with control group, the number of liquefactiveembryos,dead ones,huge ones and hypoevolutism ones were more than GDM group, thedifference was statistically significant, P<0.01. The average body weight of GDM group embryoswas significantly lower than the control group in pregnant12,15,19days, the difference wasstatistically significant, P <0.01. Compared with the control group, fetal rat huge growth andretardation occurs more than GDM group, difference was statistically significant, P<0.01.Thereare more at embryo heart development defect nurnbers at GDM group in pregnant12,15,19daysthan that of the control group,there exists obvious statistics difference, P<0.01. Major includingcaul-conus development malformation,ventricular septal defect(VSD)and myocardialhypertrophy,abnormal enlargement of the heart chamber,and so on.3. The expression of TBX-5protein: The expression of TBX-5protein in myocardial cells werestronger on the pregnant days of12,15, and19in GDM group, the difference was significantcompared with control group, P<0.01. The expression of TBX-5in the heart of control group andGDM group on the pregnant days of12were significantly higher than the days of15and19, P<0.01. The expression of TBX-5were no singnificant differences between the pregnant days of15and19in control group and GDM group, P>0.05.CONCLUSIONS: Maternal gestational diabetes can cause abnormal fetal heart development.TBX-5protein in gestational diabetes pregnancy12,15,19days of fetal cardiac expression weresignificantly increased, suggesting that TBX-5may be involved in the process caused byabnormal heart development in gestational diabetes.
